Abstract

TRAIL R1 (Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand Receptor 1) merupakan death domain (DD) yang mengandung protein transmembran tipe I yang memediasi terjadinya apoptosis pada sel kanker yang berproliferasi. Daun kelor ( Moringa oleifera ) mengandung flavonoid (terutama quercetin) yang diharapkan dapat mengembalikan sensitivitas sel-sel kanker terhadap penginduksi apoptosis. Penelitian ini bertujuan untuk membuktikan kemampuan ekstrak methanol daun kelor dalam meningkatkan ekspresi TRAIL R1 jaringan hepar yang diinduksi 7,12-Dimethylbenz(a)anthracene (DMBA). Pada penelitian ini digunakan rancangan true experimental pada 5 kelompok tikus. Masing-masing kelompok terdiri dari 6 tikus. Selama 104 hari kelompok pertama (kontrol negatif) diberi diet normal saja. Kelompok kedua (kontrol positif) tikus diberi diet normal dan DMBA 10 mg/hari selama 44 hari, kemudian dilanjutkan dengan pemberian diet normal saja selama 60 hari. Sedangkan kelompok III, IV, dan V diberi diet normal dan DMBA 10 mg/hari selama 44 hari, kemudian dilanjutkan dengan pemberian diet normal dan ekstrak methanol kelor dengan dosis yang berbeda (20, 40, 80 mg/ml/hari) selama 60 hari. Pengukuran ekspresi TRAIL R1 jaringan hepar pada penelitian ini dilakukan dengan metode immunohistochemistry . Analisis data dengan metode one way ANOVA menunjukkan ekspresi TRAIL R1 pada pemberian kelor dosis 20mg/ml/hari berbeda secara bermakna dibanding kontrol positif. Analisis data kemudian dilanjutkan dengan uji Korelasi Pearson . Kesimpulan penelitian ini adalah bahwa ekstrak methanol daun kelor ( Moringa oleifera ) mampu meningkatkan ekspresi TRAIL R1 jaringan hepar pada tikus Wistar yang diinduksi 7,12-Dimethylbenz(a)anthracene (DMBA). Dosis efektif adalah 40 mg/ml/hari.

English Abstract

TRAIL R1 (Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand Receptor 1) is a death domain (DD) containing type I transmembrane protein which mediates apoptosis toward cancer cell proliferation. Drumstick ( Moringa oleifera ) leaves contain flavonoid (mostly quercetin) that is expected to return the sensitivity of cancer cells toward apoptosis inducer. This study aims to prove the ability of methanol extract of drumstick leaves in increased TRAIL R1 expression in liver tissue induced by 7 ,12-Dimethylbenz(a)anthracene (DMBA) . This study used true experimental design in 5 groups of mice. Each group consisted of 6 mice. For 104 days, the first group (negative control) was given a normal diet alone. The second group (positive control) mice were given a normal diet and DMBA 10 mg/day for 44 days, then were continued with normal diet alone for 60 days. While the group III, IV and V were given a normal diet and DMBA 10 mg/day for 44 days, and then were continued with methanol extract of Moringa oleifera leaves with different doses (20, 40, 80 mg/ml/day) for 60 days. Measuring of liver tissue TRAIL R1 expression in this study was done by immunohistochemistry method. Data analysis by one-way ANOVA method showed that administration of methanol extract of drumstick leaves caused significant difference in TRAIL R1 expression compared to positive control group. Data analysis then was followed by Pearson Correlation Test. The conclusion of this study is methanol extract of drumstick ( Moringa oleifera ) can enhance TRAIL R1 expression in the liver tissue of Wistar mice that were induced by 7,12-Dimethylbenz(a)anthracene ( DMBA). Effective dose is 40 mg/ml/day .
