Abstract
Insisi gingiva merupakan suatu tindakan pemotongan jaringan pada gingiva yang menyebabkan terjadinya luka pada gingiva. Chitosan merupakan suatu zat yang dapat mempercepat proses penyembuhan luka. Penelitian ini bertujuan untuk membuktikan bahwa pemberian chitosan topikal terhadap angiogenesis pada proses penyembuhan pasca insisi gingiva pada tikus (Rattus novergicus) Galur Wistar. Tikus dibagi 5 kelompok, kelompok, kontrol negatif dan positif, serta 3 kelompok perlakuan (pemberian gel chitosan dengan konsentrasi berbeda). Insisi dilakukan diantara kedua insisivus rahang bawah lalu diberikan perlakuan sesuai kelompok. Tikus dibedah pada hari ke tujuh. Jumlah pembuluh darah dihitung pada preparat dengan perwarnaan HE, diamati menggunakan mikroskop digital Olympus dengan perbesaran 400x. Hasil didapatkan jumlah pembuluh darah yang berbeda nyata antara kelompok perlakuan dengan kelompok kontrol positif (p<0,05) kurang lebih 8,30 per lapang pandang; kontrol positif 5,51 per lapang pandang; perlakuan gel chitosan topikal 12,5% 10,58 per lapang pandang; perlakuan gel chitosan topikal 25% 12,28 per lapang pandang; perlakuan gel chitosan topikal 50% 16,73 per lapang pandang. Uji normalitas data nilai signifikansi 0,200 sehingga data berdistribusi normal. Uji homogenitas dengan Levene’s Test nilai signifikansi 0,79 sehingga data tersebut homogen. Uji korelasi pearson didapat r=(-0,416) menunjukan terdapat korelasi moderat antara konsentrasi gel chitosan dengan jumlah pembuluh darah. Hasil yang dapat diambil dari penelitian ini adalah pemberian gel chitosan berpengaruh terhadap angiogenesis pada proses penyembuhan pasca insisi gingiva pada tikus (Rattus novergicus) Galur Wistar.
English Abstract
A gingival incision is an act of cutting tissue in the gingiva causing injury to the gingiva. Chitosan is a substance that can accelerate the process of wound healing. This study is aimed to prove that topical chitosan administration influences angiogenesis in post-incisional gingival healing process in rats (Rattus novergicus) Galur Wistar. Rats were divided into 5 groups, negative and positive controls, and 3 treatment groups (giving chitosan gel with different concentrations). The incision was performed between the lower mandibular incisors and the treatment was given as it was needed by each group. Rats were dissected on the seventh day. The number of blood vessels were counted on preparations with HE staining was observed using an Olympus digital microscope with 400 × magnification. The result showed that the number of blood vessels was significantly different between the treatment group and the positive control group (p <0.05) about 8.30 per field of view; positive control 5, 51 per field of view; topical chitosan gel treatment of 12.5% 10.58 per field of view; topical chitosan gel treatment 25% 12,28 per field view; treatment of topical chitosan gel 50% 16,73 per field of view. The normality test data value significance of 0.200 so that the data is normally distributed. Test homogeneity with Levene's Test significance value of 0.79 so that the data is homogeneous. Pearson correlation test obtained r = (- 0.416) showed a moderate correlation between chitosan gel concentration with the number of blood vessels. The results of the study is that giving chitosan gel influences angiogenesis on healing process after gingival incision in rats (Rattus novergicus) Galur Wistar.
