Indonesian Abstract

Latar Belakang. Chronic Myeloid Leukemia (CML), adalah gangguan myeloproliferatif yang ditandai adanya peningkatan proliferatif dari sel granulositik tanpa kehilangan fungsi diferensiasi selnya, yang dikenali dengan ditemukannya kromosom Philadelphia atau produk molekular genetik protein fusi yaitu BCR/ABL. Leukositosis dan trombositosis sering ditemukan pada pasien yang didiagnosa CML. Penelitian ini bertujuan untuk mengetahui adanya korelasi jumlah ekspresi BCR-ABL terhadap jumlah leukosit dan trombosit pada pasien CML. Metode Penelitian. Penelitian crosssectional study dengan pengambilan sampel secara konsekutif dari bulan September 2016-Februari 2017. Subyek 30 individu dengan qRT-PCR BCR-ABL positif dibagi menjadi kelompok jumlah leukosit ≤20.000/μL dan >20.000/μL, kelompok jumlah trombosit ≤400.000/μL dan >400.000/μL. Hasil Penelitian. Terdapat perbedaan bermakna rerata jumlah BCR—ABL pada kelompok leukosit >20.000/μL (2581533,33 copies/mL±1709921,88) yaitu yang lebih tinggi dibanding dengan kelompok jumlah leukosit ≤20.000/μL (282572,98/μL±66938,28). Namun tidak terdapat perbedaan bermakna rerata jumlah BCR-ABL kelompok subyek dengan jumlah trombosit ≤400.000/μL (1336845,212/μL±1757728,86) dibandingkan dengan jumlah leukosit >400.000/μL (1693875/μL±1464136,94). Terdapat korelasi positif dengan kekuatan korelasi kuat antara jumlah BCR-ABL dengan jumlah leukosit yaitu r=0,701, p< 0,01, dan tidak terdapat korelasi antara BCR-ABL dengan jumlah trombosit r=0,096, p=0,613. Kesimpulan. Perbedaan yang bermakna didapatkan pada jumlah BCR-ABL positif terhadap jumlah leukosit Korelasi positif yang kuat didapatkan pada jumlah BCR-ABL terhadap jumlah leukosit.

English Abstract

Background. Chronic Myeloid Leukemia (CML), is a myeloproliferative disorder characterized by a proliferative increase of granulocytic cells without loss of cell differentiation function, which is identified by the discovery of Philadelphia chromosomes or molecular genetic products of fusion protein BCR/ABL. Leukocytosis and thrombocytosis are common in patients diagnosed with CML. This study aims to determine the correlation of the number of BCR-ABL expression on the number of leukocytes and platelets in CML patients. Research methods. A cross-sectional study study with a consecutive sampling from September 2016-February 2017. The subjects of 30 individuals with qRT-PCR BCR-ABL were positively divided into groups of leukocyte counts ≤20.000/μL and >20,000/μL, platelet count groups ≤400.000/μL and >400,000/μL. Research result. There was a significant difference in the mean number of BCR-ABL in the leukocyte group >20.000/μL (2581533,33 copies/mL±1709921,88), which was higher than the leukocyte count group ≤20.000/μL (282572,98/μL±66938,28). However, there was no significant difference in mean number of BCR-ABL group subjects with platelet count ≤400.000/μL (1336845,212/μL±1757728,86) compared with leukocyte counts >400,000/μL (1693875/μL±1464136,94). There was a positive correlation with a strong correlation strength between the number of BCR-ABL with the number of leukocytes that is r = 0.701, p <0.01, and there was no correlation between BCR-ABL with platelet count r = 0.096, p = 0.613. Conclusion. A significant difference was found in the positive number of BCR-ABL on the number of leukocytes A strong positive correlation was obtained in the number of BCR-ABL on the number of leukocytes.
