Indonesian Abstract

Tomat merupakan salah satu buah yang mengandung antioksidan dalam jumlah besar. Salah satu antioksidan yang banyak di tomat adalah likopen. Penggunaan antioksidan yang tinggi dapat menimbulkan aktivitas pro-oksidan yang menimbulkan efek toksik pada hati. Penelitian ini dilakukan untuk mengetahui efek paparan ekstrak kulit tomat dosis 2000 mg/KgBB terhadap kadar SGOT dan SGPT tikus Rattus norvegicus strain wistar betina dan jantan. Penelitian ini merupakan penelitian true experimental design dengan randomized only post test controlled group design secara in vivo pada 8 kelompok tikus Rattus norvegicus betina dan jantan yang masing-masing kelompok terdiri dari 3 tikus. Ekstrak kulit tomat diperoleh dengan metode maserasi dengan pelarut etanol 98%. Parameter yang diamati adalah aktivitas enzim SGOT dan SGPT dalam 24 jam dan 14 hari. Hasil pengamatan pada paparan 24 jam dan 14 hari tidak didapatkan tanda-tanda toksik yaitu tidak terdapat perubahan kulit dan bulu, mata dan membran mukosa, tremor, diare, letargi, dan koma. Hasil analisis kadar SGOT serum menggunakan uji One-Way ANOVA antar kelompok tikus betina kontrol dan perlakuan 24 jam dan 14 hari memiliki nilai signifikansi p = 0,983 dan p = 0,787 yang artinya tidak didapatkan perbedaan yang bermakna antar kelompok. Pada kelompok tikus jantan kontrol dan perlakuan 24 jam dan 14 hari memiliki nilai signifikansi p = 1,000 dan p = 1,000 menunjukkan tidak didapatkan perbedaan yang bermakna antar kelompok. Hasil uji One-Way ANOVA untuk kadar SGPT serum antar kelompok tikus betina maupun jantan kontrol dan perlakuan 24 jam dan 14 hari juga menunjukkan bahwa tidak didapatkan perbedaan yang bermakna antar kelompok dengan nilai signifikansi p = 0,812 dan p = 1,000 untuk tikus betina dan nilai p = 0,986 dan p = 1,000 untuk tikus jantan. Dapat disimpulkan bahwa paparan akut ekstrak kulit tomat dosis 2000 mg/KgBB belum menimbulkan kerusakan hati tikus dilihat dari kadar SGOT serum dan SGPT serum antar kelompok tikus betina dan jantan.

English Abstract

Tomatoes are fruit that containantioxidants in large quantities. One of the many antioxidants in tomatoes is lycopene. Using of high antioxidants can lead to pro-oxidant activity that cause toxic effects on the liver. This study was conducted to determine the effect of tomato skin extract dose 2000 mg / KgBB on levels of SGOT and SGPT rat Rattus norvegicus female and male wistar strains. This research is true experimental design with randomized only post test controlled group design in vivo on 8 groups of female and male Rattus norvegicus rats, each group consisting of 3 mice. Tomato skin extract was obtained by maceration method with 98% ethanol solvent. The parameters observed were SGOT and SGPT enzyme activity in 24 hours and 14 days. Results of observation at 24 hours and 14 days exposure did not show any signs of toxic there are no change of skin and fur, eyes and mucous membranes, tremors, diarrhea, lethargy, and coma. Results of serum SGOT analysis using One-Way ANOVA test between control female and 24 hours and 14 days had significance value of p = 0,983 and p = 0,787, meaning that there was no significant difference between groups. In the control and treatment rats group 24 hours and 14 days had significance value p = 1,000 and p = 1,000 showed no significant difference between groups. One-Way ANOVA test results for serum SGPT levels between female and male control groups and 24-hour and 14-day controls also showed no significant differences between groups with significance values of p = 0.812 and p = 1,000 for female rats and p-values = 0.986 and p = 1,000 for male rats. It can be concluded that acute exposure of tomato skin extract dose 2000 mg / KgBB has not caused rat liver damage seen from serum SGOT level and serum SGPT among groups of female and male rats.
