Indonesian Abstract

Diabates meIIitus tipe 2 (DM 2) merupakan kondisi yang disebabkan oIeh berkurangnya insulin di dalam tubuh atau resistensi insuIin. Resistensi insuIin dapat menyebabkan ketidakseimbangan antara PPAR α dan PPAR ɣ yang mengakibatkan terjadinya Non-aIcohoIic Fatty Liver Disease (NAFLD) atau disebut juga dengan perIemakan. Pemberian vitamin A dapat menurunkan jumIah perIemakan seI hepatosit secara signifikan dikarenakan fungsi dari vitamin A yang mampu menurunkan penumpukan Iemak, meningkatkan hidroIisis trigliserida daIam hepar, dan meningkatkan oksidasi Iemak melalui aktivasi retinoic X receptor (RXR). Penelitian ini bertujuan untuk mengetahui efek pemberian vitamin A terhadap perIemakan sel hepatosit pada tikus putih (Rattus norvegicus) galur Wistar jantan model DM 2. Penelitian menggunakan metode randomized post-test experimental secara in vivo di Iaboratorium. SampeI dibagi menjadi 5 keIompok, yang terdiri atas kontroI negatif, kontroI positif, perlakuan 1 (vitamin A dosis 50 mg/kgBB), perIakuan 2 (vitamin A dosis 100 mg/kgBB), dan perIakuan 3 (vitamin A dosis 150 mg/kgBB). Pada awaI perIakuan, pemberian diet tinggi Iemak dan injeksi STZ diIakukan pada keIompok kontroI positif dan ketiga keIompok perIakuan. Pemberian vitamin A diIakukan seteIah tikus dinyatakan DM 2 dan berlangsung seIama empat minggu perIakuan. SeIanjutnya, diIakukan pembedahan pengambiIan organ hepar untuk diproses secara histopatoIogis guna pengamatan perIemakan sel hepatosit. HasiI peneIitian menunjukkan bahwa terdapat perbedaan yang bermakna antara jumIah perIemakan pada semua keIompok yang dibandingkan (ANOVA p=0.000). Berdasarkan peneIitian ini disimpuIkan bahwa vitamin A memiIki efek untuk menurunkan jumIah perIemakan sel hepatosit.

English Abstract

Diabetes mellitus type 2 (DM 2) is a condition that caused by deficiency of insulin in the body or insulin resistance. Insulin resistance can cause an imbalance between PPAR α and PPAR ɣ resulting in the occurrence of Non-aIcohoIic Fatty Liver Disease (NAFLD) or also called fatty liver. Vitamin A can decrease the number of fatty liver in hepatocytes significantly due to the function of vitamin A that can decrease the fat accumulation, increase triglycerides hydrolysis in the liver, and increase the fat oxidation through the activation of retinoic X receptor (RXR). This study is aimed to know the effect of vitamin A on the fatty liver of rats’ hepatocytes (Rattus norvegicus) male Wistar strains of DM 2. This study used in vivo randomized post-test experimental method in the laboratory. The samples were divided into 5 groups, consisting of negative control, positive control, treatment 1 (vitamin A 50 mg / kgBB), treatment 2 (vitamin A 100 mg / kgBB), and treatment 3 (vitamin A 150 mg / kgBB). On the beginning of the treatment, high-fat diet and STZ injection were given to the positive control group and the three groups of treatment. Vitamin A was given after the rats were stated DM 2 and lasted for four weeks treatment. Furthermore, surgical removal of liver is done, to be processed histopathologically for observation of fatty liver on hepatocytes. The results showed that there was a significant difference between the number of fatty liver in all of the compared groups (ANOVA p = 0.000). Based on this study it is concluded that vitamin A has the effect to decrease the number of fatty liver on hepatocytes.
