Indonesian Abstract

Diabetes Melitus tipe 2 adalah penyakit metabolik yang ditandai dengan hiperglikemia disebabkan oleh resistensi insulin. Salah satu komplikasi yang ditimbulkan yaitu kerusakan jaringan pada otak. Hiperglikemia memiliki peran dalam pembentukan Reactive Oxygen Species (ROS) yang dapat meningkatkan modifikasi DNA, lemak, dan protein diberbagai jaringan. Terjadinya peningkatan stres oksidatif menyebabkan degenerasi sel saraf otak termasuk sel piramidal cortex cerebri. Kulit tomat banyak mengandung antioksidan seperti likopen, β- karoten, flavonoid, fenolat dan asam askorbat, diharapkan mampu menurunkan kadar radikal bebas sehingga mencegah progresivitas komplikasi Diabetes Melitus tipe 2. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kulit tomat terhadap degenerasi sel piramidal cortex cerebri tikus (Rattus norvegicus). Penelitian ini menggunakan true experimental design. Sejumlah 20 ekor tikus galur wistar terbagi menjadi lima kelompok, yaitu: kelompok normal, kelompok DM tipe 2, dan kelompok perlakuan ekstrak kulit tomat dengan dosis 50mg/kgBB, 100mg/kgBB, dan 150mg/kgBB secara peroral selama 4 minggu. Penghitungan degenerasi sel piramidal cortex cerebri secara histopatologi dalam 10 lapang pandang menggunakan aplikasi Master Scan Dot Slide dengan perbesaran 400x. Analisis data menggunakan uji One Way ANOVA, uji Post-hoc dan uji Korelasi. Hasil degenerasi sel piramidal pada kelompok kontrol normal, kontrol DM tipe 2, kelompok perlakuan ekstrak kulit tomat dengan dosis 50mg/kgBB, 100mg/kgBB, dan 150mg/kgBB secara berturut-turut adalah (dalam satuan %) 18.503, 72.227, 37.418, 36.649, 35.813. Ada penurunan yang signifikan pada kelompok perlakuan ekstrak kulit tomat terhadap kelompok DM tipe 2 (p=0.000). Uji Korelasi menunjukkan ada hubungan yang signifikan antara pemberian dosis ekstrak kulit tomat dengan degenerasi sel piramidal cortex cerebri (p=0.004). Kesimpulannya pemberian ekstrak kulit tomat dapat menurunkan degenerasi sel piramidal cortex cerebri.

English Abstract

Diabetes Mellitus type 2 is a metabolic disease that marked with hyperglycemia that caused by insulin resistance. One of the complications of it is a damage of the tissue on the brain. Hyperglycemia has a lead on the formation of Reactive Oxygen Species (ROS) that can increase modification of DNA, fat, protein on the various tissues. As the oxidative stress occur, it causing brain nerve cell degeneration including pyramidal cell cortex cerebri. Tomato skin’s that contain a lot of antioxidants such as lycopene, β-carotene, flavonoid, phenolic and ascorbic acid, expected to be able to decrease the amount of free radical so it can prevent the complication progressivity of Diabetes Mellitus type 2. This researchers purpose is to find out the influence of tomato skin's extract toward degeneration of pyramidal cell cortex cerebri to rat (Rattus norvegicus). This research used true experimental design. Using 20 wistar strain rat’s which is divided by five groups, i.e: normal group, DM Type 2 group, and tomato skin's extract with dosage 50mg/kgBW, 100 mg/kgBW, and 150mg/kgBW group treatment is given orally for 4 weeks. Calculation of pyramidal cell cortex cerebri degeneration in histopathology in 10 field of view using Master Scan Slide application with 400x magnification. Data analysis used One Way ANOVA test, Post-hoc test and Correlation test. The result of pyramidal cell degeneration in normal control group, type 2 DM control, treatment group of tomato skin’s extract at doses of 50mg/kgBW, 100mg/kgBW, and 150mg/kgBW respectively were (in%) 18.503, 72.227, 37.418, 36.649, 35.813. There was a significant decrease in treatment group of tomato skin‘s extract to DM type 2 group (p = 0.000). Correlation test showed significant correlation between dosage of tomato skin’s extract with pyramidal cell cortex cerebri degeneration (p = 0.004). The conclusion is giving tomato skin's extract can decrease degeneration of pyramidal cell cortex cerebri.
