Indonesian Abstract

Hiperkolesterolemia adalah suatu kondisi dimana meningkatnya konsentrasi kolesterol dalam darah yang melebihi nilai normal. Penggunaan obat hiperkolesterolemia atau obat sintetis jangka panjang akan menimbulkan efek samping, sehingga diperlukan adanya alternatif pencegahan yang berasal dari bahan alami. Arang aktif dapat menyerap ribuan kali beratnya sendiri zat-zat berupa gas, logam berat, racun-racun dan bahan-bahan kimia lain, sehingga merupakan bahan yang efektif dan tidak berbahaya. Penelitian ini bersifat true experimental menggunakan Rancangan Acak Lengkap (RAL). Hewan model menggunakan tikus putih (Rattus norvegicus) strain Wistar jantan berumur 8 – 12 minggu, dengan berat badan antara 100-150 g diadaptasikan selama 7 hari dan dibagi dalam enam kelompok perlakuan. Kelompok negatif, positif, kelompok perlakuan 1, 2, 3 dan 4. Pemberian diet hiperkolesterolemia menggunakan bahan asam kholat 0,1%, minyak babi 10%, dan kuning telur puyuh segar 5 % dengan total pemberian 3,02 gr/2 mL selama 14 hari. Suplementasi menggunakan arang aktif dengan dosis masing-masing 0,34 g/150gBB, 0,68 g/150gBB, 1,02 g/150gBB dan perlakuan 4 dengan dosis 0,68 g/150gBB tanpa pemberian diet hiperkolesterolemia, pemberian dengan sonde lambung selama 14 hari. Pengukuran kadar MDA menggunakan uji TBA, analisa data dilakukan secara kuantitatif menggunakan uji One Way ANOVA, α = 5% dan uji tukey (p<0,01). Hasil gambaran histopatologi organ jejunum dianalisa secara deskriptif. Hasil analisis statistik kadar MDA, pada perlakuan 2 dengan dosis arang aktif 0,68 g/150gBB memberikan pengaruh terbaik terhadap pencegahan peningkatan kadar MDA dan mencegah kerusakan tunika mukosa. Kesimpulan dari penelitian ini adalah arang aktif dapat digunakan dalam mencegah hiperkolesterolemia dengan mencegah kenaikan kadar MDA dan mencegah kerusakan gambaran histopatologi jejunum.

English Abstract

Hypercholesterolemia is a condition in which blood cholesterol level increased abnormally. The use of hypercholesterolemia drugs or long-term synthetic drugs result in side-effects, thus a prevention alternative derived from natural sources is necessary. Activated charcoal has the ability to absorb gas, heavy metals, toxins, and other chemical substances, thus making it an effective and harmless alternative to the aforementioned drugs. This study used complete random design (RAL) method which has a true experimental nature. Model organisms used in this experiment are white mice Wistar strain (Rattus norvegicus) aged 8 to 12 weeks, with body mass ranging from 100 to 150 g, adapted for 7 days and separated into six variable groups: negatives, positives, treatment 1, 2, 3, and 4. Hypercholesterolemia diet consisting of 0.1% cholate acid, 10% lard, and 5% freshly quail egg yolks were each given to the mice in the amount of 3.02 gr/2 mL for 14 days. Activated charcoal supplement was given in the dose of 0.34 g/150 g body mass (BM), 0.68 g/150gBM, and 1.02 g/150gBM for treatment 1, 2, and 3, respectively. Meanwhile, treatment 4 is dosed 0.68 g/150gBM with no hypercholesterolemia diet treatment. These supplementations were given for 14 days with the help of feeding tube. Malondialdehyde (MDA) level was measured using thiobarbituric acid (TBA) test, in which data analysis was performed quantitatively using ANOVA one way test, α = 5%, and tukey test, p<0.01. Jejunum histopathology results were analyzed descriptively. Based on the statistical analysis of MDA level test results, 0.68g/150gBM dose of activated charcoal on the treatment 2 gives the best result in preventing the increase of MDA level and preventing damage to mucosal tunica. In conclusion, activated charcoal can aid in preventing hypercholesterolemia by reducing the level of MDA and preventing damage in jejunum histopathological features.
