Indonesian Abstract

Fibrosis hepar termasuk salah satu penyakit sistemik yang umum terjadi pada hewan kecil dan belum ditemukan terapi yang efektif. Ekstrak gambir (Uncaria gambir Roxb.) memiliki kandungan flavonoid diharapkan dapat digunakan untuk pencegahan penyakit ini. Penelitian ini dilakukan untuk mengetahui pengaruh preventif ekstrak gambir (Uncaria gambir Roxb.) berdasarkan aktivitas enzim Superoksida Dismutase (SOD) dan banyaknya jaringan ikat pada histopatologi hepar. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan hewan coba tikus (Rattus norvegicus) jantan berumur 8-12 minggu dengan berat badan 150-200 gram. Tikus dibagi menjadi 5 kelompok yaitu, kontrol negatif (tanpa induksi CCl4 dan preventif), kontrol positif (diinduksi CCl4), kelompok preventif dosis 130 mg/kg BB, 260 mg/kg BB, dan 390 mg/kg BB. Pengukuran kadar SOD menggunakan spektofotometer dianalisis menggunakan ragam one-way ANOVA dan dilanjutkan dengan uji Tukey α=5%, serta pengamatan histopatologi hepar dengan pewarnaan Masson Trichome dijelaskan secara deskriptif. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak gambir dapat mencegah fibrosis hepar berdasarkan kenaikan kadar SOD dan berkurangnya jaringan ikat yang terbentuk pada histopatologi hepar. Kesimpulan dari penelitian ini yaitu dosis ekstrak gambir 390 mg/kg BB merupakan dosis terbaik sebagai preventif fibrosis hepar.

English Abstract

Liver fibrosis is one of systemic diseases that are common in small animals that currently have not found effective therapy for this disease. Gambier extract (Uncaria gambir Roxb.) that contain flavonoids have antioxidant activity expected to be used for prevention of this disease. This research was conducted to determine the effect of preventive of gambir extract (Uncaria gambir Roxb.) based on the activity of the enzyme Superoxide Dismutase (SOD) and the amount of connective tissue in liver’s histopathology. This study used a Randomized Complete Design (RAL), the experimental animals used were rats (Rattus norvegicus) males aged 8-12 weeks weighing 150-200 grams. Rats were divided into five groups: the negative control (without CCl4 induction and preventive), positive control (CCl4-induced), group preventive doses of 130 mg/kg, 260 mg/kg, and 390 mg/kg. Measurement of the levels of SOD using spectrophotometer which is analyzed using a variety of a one-way ANOVA and Tukey test with α=5%, and also observed liver’s histopathology with Masson Trichome staining described are descriptively. The results of this study indicate that the giving of gambir extract could be prevent liver fibrosis based on examination of increase the levels of SOD and observations of liver’s histopathology with less of connective tissue formed. The conclusions of this study, namely dose of gambir extract about 390 mg/kg BB is prefer dose as preventive of liver fibrosis.
