Indonesian Abstract

Pencemaran lingkungan terjadi akibat terganggunya suatu lingkungan sehingga mengakibatkan ketidakseimbangan ekosistem akibat perubahan beberapa faktor yang ada di dalamnya. Pencemaran perairan di kawasan pesisir pada umumnya adalah pencemaran logam berat yang bersumber dari aktivitas manusia di sekitar pesisir pantai. Tiram Crassostrea iredalei dapat dijadikan sebagai biomonitoring karena kemampuannya sebagai filter feeder dan hidupnya yang menetap sehingga mampu mengakumulasi logam berat. Penelitian ini menggunakan metode deskriptif, dimana sampel tiram diambil dari tiga stasiun yaitu di dermaga pelabuhan, tempat pendaratan ikan dan kawasan mangrove BeeJay Bakau Resort (BJBR) Probolinggo. Analisis nilai densitas dan intensitas metallothionein digunakan metode Imunohistokimia, untuk perhitungan nilai densitas dan intensitas digunakan program imageJ, sedangkan untuk analisis kadar logam berat Pb, Cd dan Hg digunakan metode Atomic Absorbsion Spectrofotometer (AAS). Hasil analisis menunjukkan kandungan logam berat Pb pada insang tiram berkisar 0,387-0,64 mg/l, Cd berkisar 0,092 -0,215 mg/l, dan Hg berkisar 0,082– 0,158 mg/l, sementara itu kandungan logam berat Pb di lambung berkisar 0,202–0,434 mg/l, Cd berkisar 0,045–0,074 mg/l, dan Hg berkisar 0,055-0,085 mg/l. Hasil analisis kadar densitas metallothionein pada insang tiram menunjukkan nilai 0,0309 MT/μm2 - 0,0477 MT/μm2, sementara itu pada lambung nilai densitas berkisar antara 0,0380 MT/μm2 – 0,0421 MT/μm2. Intensitas metallothionein pada insang tiram berkisar antara 39.581 – 68.547 pixel, sedangkan nilai intensitas pada lambung berkisar antara 61.951 – 82.648 pixel. Uji analisis regresi dengan SPSS menunjukkan adanya hubungan yang sangat kuat antara logam berat terhadap densitas dan intensitas MT pada insang dan lambung tiram dengan koefisien korelasi (r) berkisar antara = 0,923 – 0,985.

English Abstract

Environmental pollution is occured when the disruption of environment is already existed and caused the ecosystems out of balance due to change of several factors that exist within it. Generally coastal pollution in the certain areas caused by heavy metal pollution which are discharged to the water by human activities around the coast. Oyster Crassostrea ireldaei can be used as a biomarker because its ability as filter feeder, and they are normally settled to accumulate heavy metals. Using descriptive method, in this research oyster samples were collected from three stations, i.e. harbor, fish landing sites and mangrove areas of BJBR Probolinggo. To analysis the density value and the intensity of MT the Immunohistochemistry method had been used, and the density value and intensity were calculated by using imageJ program. Meanwhile, the heavy metal Pb, Cd and Hg were analysed by using Atomic Absorbsion Spectrofotometer (AAS) method. The results revealed that heavy metals were accumulated in the gills of oysters, where Pb concentration was within the range of 0.387 to 0.64 mg/l, Cd ranged from 0.215 - 0.092 mg/l, and Hg range from 0,082 - 0.158 mg/l. Meanwhile in the stomach of oyster the Pb content ranged from 0.202 - 0.434 mg/l, Cd ranged from 0.045 - 0.074 mg/l, and Hg ranged from 0.055 - 0.085 mg/l. Analysis of the density levels of metallothionein in oyster gills indicated the value of 0,0309 MT/μm2 to 0,0477 MT/μm2, while on the stomach the density value ranged from 0.0380 MT/μm2 to 0,0421 MT/μm2. Further, metallothionein intensity in oyster gills ranged from 39,581 to 68,547 pixels, and on the stomach were found between 61,951 - 82,648 pixels. Using SPSS for regression analysis, it was found that there was a very strong relationship of correlation between heavy metal content with density and intensity of MT on gill and stomach oyster as indicated by correlation coefficient (r) value which ranged from 0,923 to 0,985.
