Indonesian Abstract

Diabetes Mellitus tipe 2 (DM tipe 2) adalah gangguan metabolisme yang ditandai dengan adanya resistensi insulin dan berkurangnya jumlah insulin dalam darah. DM tipe 2 dapat menyebabkan komplikasi ke berbagai organ akibat keadaan hiperglikemia yang muncul dan menyebabkan kerusakan pada beberapa organ. Latihan fisik merupakan salah satu cara yang diharapkan dapat mengurangi kerusakan dan gangguan pada organ. Penelitian ini bertujuan untuk mengetahui pengaruh latihan fisik (treadmill) terhadap kadar Malondialdehyde (MDA) dan kadar Superoksida dismutase (SOD) pada tikus (Rattus norvegicus) model DM tipe 2. Tikus model DM tipe 2 dihasilkan dengan induksi Streptozotocin 30 mg/Kg BB multiple low-dose secara intraperitoneal diberikan satukali dalam satu hari pada hari ke 29 dan 36, dan pemberian 40 gram/ekor/hari pakan tinggi lemak (High Fat-Diet) selama 63 hari. Penelitian ini menggunakan empat kelompok tikus, yaitu kelompok normal sedentari, normal latihan fisik, DM tipe 2 sedentari dan DM tipe 2 latihan fisik. Latihan fisik dilaksanakan 6 hari dalam 1 minggu dan kecepatan bertahap. Kecepatan dan durasi latihan fisik setiap 2 minggu mengalami peningkatan, dimulai dari kecepatan 10 meter/menit selama 10 menit, 14 meter/menit selama 22 menit, 18 meter/menit selama 34 menit, 22 meter/menit selama 46 menit, dan 27 meter/menit selama 60 menit. Kadar MDA dan aktivitas SOD pada organ lambung diukur menggunakan spektofotometer selanjutnya dianalisis secara kuantitatif menggunakan uji one way ANOVA α=5% kemudian dilanjutkan dengan Uji BNJ (beda nyata jujur). Hasil uji ANOVA menunjukan latihan fisik dapat meningkatkan kadar MDA secara signifikan (p<0.005) dan latihan fisik secara signifikan dapat menurunkan kadar SOD pada hewan model DMT2 induksi STZ dan HFD.

English Abstract

Diabetes mellitus type 2 (DM type 2) is a metabolism disorder which is characterized by the presence of insulin resistance and reduced amount of insulin in the blood. DM type 2 can cause complication to any organ for the appearing hyperglicemia condition and the increasing of Reactive Oxygen Species (ROS) production. Physical exercise is one of many DM type 2 management which is expected to reduces any damages and disorders to the affected tissues. The purpose of this research was to investigated the effect of physical exercise (treadmill) against levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in gastrics of rats (rattus norvegicus) model with diabetes mellitus type 2. DM type 2 rats model was generated with the induction of Streptozotocin 30mg/Kg BB multiple low-dose by intraperitoneal route and feed with 40gram/rat/day high fat diet for 63 days. This research used 4 groups of rats, which were normal sedentary group, normal with physical exercise group, DM type 2 sedentary group and DM type 2 with physical exercise. The speed and duration of physical exercise every 2 weeks increases, starting from 10 meters/minute for 10 minutes, 14 meters/minutes for 22 minutes, 18 meters/min for 34 minutes, 22 meters/minute for 46 minutes and 27 meters/minute for 60 minutes. Levels of MDA was measured with Thiobarbituric Acid Ractive Substrate (TBARS) and levels of SOD was measured by using spectrophotometer which were then analysed quantitatively using one way ANOVA test at α=5%. The ANOVA test results showed that physical exercise could significantly increase MDA levels (p <0.005) and physical exercise significantly reduced SOD levels in animal models of DM type 2 induction STZ and HFD.
