Indonesian Abstract

Glutathion dimanfaatkan dalam industri makanan, farmasi, dan kosmetik. Rekayasa metabolisme melalui penyisipan gen gshI ke dalam vektor ekspresi plasmid pESC-TRP serta peningkatan biomassa yeast melalui perpanjangan waktu panen menjadi upaya yang efektif dalam meningkatkan produksi GSH. Tujuan dari penelitian ini adalah menganalisis pengaruh waktu pemanenan glutathion pada jam ke-44 dan ke-48 terhadap produksi glutathion, biomassa dan ukuran sel Saccharomyces cerevisiae strain wild type dan mutan gshI. Metode penelitian meliputi preparasi media pertumbuhan, pembuatan kurva standard dan kurva pertumbuhan S.cerevisiae strain wild type dan mutan gshI, pengukuran glutathion intraselular, pengukuran biomassa, dan penentuan ukuran sel. Hasil penelitian ini menunjukkan bahwa tidak terdapat perbedaan yang signifikan pada kadar glutathion, biomassa sel, densitas sel, dan ukuran sel Saccharomyces cerevisiae wild type dan overekspresi gshI perlakuan kontrol dan perlakuan albumin ikan yang masing – masing dipanen pada jam ke-44 dan jam ke-48 inkubasi. Rata – rata kadar glutathion kedua strain stabil pada 1,5 – 2,5 mg/L. Total biomassa kedua strain menurun di jam ke-48 inkubasi yaitu sebesar 13,33 % pada strain wild type dan 14,08 % pada strain overekspresi gshI. Densitas S.cerevisiae wild type dan overekspresi gshI stabil pada 8,4 – 10 CFU/ml. Panjang sel Saccharomyces cerevisiae wild type dan overekspresi gshI setara pada tiap perlakuan, yaitu antara 5 – 6 μm.

English Abstract

Glutathion is widely used in the food, pharmaceutical, and cosmetic industries. Metabolic engineering through the insertion of the gshI gene into the pESC-TRP plasmid expression vector and increasing yeast biomass through the extension of the harvest time had been effective to increase GSH production. This study aims to analyze the effect of glutathione harvesting time at the 44th and 48th hours towards production of glutathione, biomass and cell size of Saccharomyces cerevisiae wild strain and gshI mutant. The methods included growth medium preparation, generation of standard curve and growth curve of S.cerevisiae wild type and gshI mutants strains, intracellular glutathione measurements, biomass measurements, and cell size determination. The results showed that there were no significant differences of glutathione production, cell biomass, cell density and cell size in control and fish albumin treatment of Saccharomyces cerevisiae wild type and gshI overexpressed harvested in 44 hour and ke-48 hour incubation time. The average of glutathion production of both strain was stable at 1,5 – 2,5 mg/L. Total biomass of both strain decreased 13,33 % in wild type strain and 14,08 % in gshI overexpressed strain in 48 hour incubation. Density of S.cerevisiae wild type and gshI overexpressed was stable at 8,4 – 10 CFU/ml. Cell length of Saccharomyces cerevisiae wild type and gshI overexpressed similar in every treatment between 5 – 6 μm.
