Indonesian Abstract

Asam laktat banyak diproduksi melalui metode fermentasi, salah satunya menggunakan bahan baku tongkol jagung dengan bantuan Rhizopus oryzae. Besarnya yield asam laktat melalui metode fermentasi dipengaruhi oleh beberapa faktor yakni konsistensi substrat dan waktu fermentasi. Oleh karena itu, diperlukan adanya kajian lebih lanjut terkait pengaruh kedua faktor tersebut terhadap pembentukan nilai yield asam laktat. Pada penelitian ini, dilakukan preparasi bahan baku melalui pengeringan menggunakan tray dryer dan reduksi ukuran tongkol jagung hingga berukuran serbuk. Kemudian dilakukan pembuatan media fermentasi dengan variasi konsistensi substrat sebanyak 10%, 15%, dan 20% (w/v). Proses fermentasi dilakukan dengan variasi lama waktu fermentasi 48, 60, dan 72 jam. Fermentasi dilakukan pada suhu 33℃ (±3 ℃), pH awal = 6 (±0,1) pada kondisi aerobik. Dilakukan pemisahan padatan dari produk fermentasi dengan cara filtrasi dan pemekatan menggunakan rotary evaporator. Pengambilan sampel dilakukan setelah fermentasi berlangsung. Kemudian dilakukan titrasi asam basa untuk mengetahui total produk asam yang dihasilkan dimana total produk asam diasumsikan sebagai total produk asam laktat. Hasil dari penelitian yaitu semakin tinggi konsistensi substrat tongkol jagung dari 10%, 15%, dan 20% (w/v) maka nilai yield asam laktat semakin rendah. Semakin lama waktu fermentasi dari 48, 60, dan 72 jam maka nilai yield asam laktat semakin meningkat. Nilai yield asam laktat tertinggi sebesar 0,78%. Nilai yield ini dihasilkan saat konsistensi substrat tongkol jagung 10% (w/v) dan lama waktu fermentasi selama 72 jam.

English Abstract

Lactic acid is widely produced by fermentation, one of the raw materials used for the fermentation is corn cob using the help of Rhizopus oryzae. Lactid acid yield from the fermentation is affected by some factors such as substrate consistency and fermentation time. Therefore, further study regarding the effect of these two factors to fermentation yield is needed. In these research, raw material is treated by drying using tray dryer and continued by reduction of corn cobs size until it becomes powder. After that, fermentation medium with substrate consistency of 10%, 15%, and 20% (w/v) are created. Fermentation time for each samples are 48, 60, and 72 hours. Fermentation temperature is kept at 30℃ (±3 ℃), starting pH = 6, and the fermentation is done aerobically. After fermentation, solids residue are separated using filtration and the filtrate is concentrated using rotary evaporator. Sample collection is done after the fermentation. To calculate the total acid product in the filtrate, acid base titration is used, where the total acid produced is assumed as total lactic acid produced. It is found that as the substrate consistency increases from 10%, 15%, and 20%, lactic acid yield decreases. Also, the longer fermentation time used, lactic acid yield also increases. It is found that maximum lactic acid yield from the fermentation is 0,78%. This data is collected from 10% substrate consistency and 72 hours of fermentation.
