Abstract
Follicle stimulating hormone receptor (FSHR) merupakan reseptor khusus dalam sel granulosa ovarium tempat follicle stimulating hormone bekerja. Infertilitas post partum merupakan kegagalan fertilisasi pada betina pasca lahir. Penelitian ini bertujuan untuk mengidentifikasi keragaman gen FSHR pada sapi perah peranakan Frisian Holstein betina dan hubungannya dengan infertilitas post partum. Sampel yang digunakan adalah 14 sampel darah sapi perah peranakan Frisian Holstein yang mengalami infertilitas post partum dan 14 sampel darah sapi perah ras yang sama, yang tidak mengalami infertilitas post partum. Ekstraksi DNA, uji kualitas, polymerase chain reaction dilakukan di laboratorium Animal Disease Diagnostic Fakultas Kedokteran Hewan Universitas Brawijaya dan uji kuantitatif di Universitas Islam Negeri Maulana Malik Ibrahim Malang. Amplifikasi fragmen gen FSHR dilakukan dengan sepasang primer forward (FSHR forward) 5’-TCCCTGCCCTTCAGTGACGAAC-3’ dan primer reverse (FSHR reverse) 5’-AGATACGCCGTCCCTTTACCT-3’. Keragaman genetik FSHR pada sapi perah peranakan Frisian Holstein betina yang mengalami infertilitas post partum dianalisis dengan metode PCR-RFLP. Hasil PCR-RFLP dianalisis secara deskriptif untuk menentukan keragaman gen FSHR. Hubungan antara keragaman gen FSHR dengan infertilitas postpartum dianalisis dengan uji korelasi Spearman menggunakan perangkat lunak Rstudio ®. Hasil uji korelasi Spearman menunjukkan bahwa jumlah potongan pita DNA antara sampel sapi perah peranakan Frisian Holstein betina yang mengalami infertilitas post partum dan yang tidak mengalami infertilitas post partum memiliki korelasi yang kuat dan berlawanan arah
English Abstract
Follicle stimulating hormone receptor (FSHR) is a special receptor which is located in granulose cell in ovary where follicle stimulating hormone (FSH) works. Post partum infertility is a fertility failure on female after parturition. This study has aim to identify FSHR gene variation on female Frisian Holstein crossbred and its correlation to post partum infertility. Samples that are used for this study are 14 blood samples from female Frisian Holstein crossbred with post partum infertility and 14 blood samples from female Frisian Holstein crossbred without post partum infertility. DNA extraction, quality test, polymerase chain reaction are done at Animal Disease Diagnostic laboratory, Faculty of Veterinary Medicine Universitas Brawijaya and quantity test is done at Universitas Islam Negeri Maulana Malik Ibrahim Malang. FSHR gene amplification is done by a pair of primer which are forward primer (FSHR forward) 5’- TCCCTGCCCTTCAGTGACGAAC-3’ and reverse primer (FSHR reverse) 5’- AGATACGCCGTCCCTTTACCT-3’. FSHR gene variation on female Frisian Holstein crossbred with post partum infertility is analyzed by PCR-RFLP method. Resulf of PCR-RFLP is analyzed descriptively to determine FSHR gene variation. Correlation between FSHR gene variation and post partum infertility is analyzed by Spearman correlation test using Rstudio ® software. Result of Spearman correlation test is there is correlation between two kind of samples
