Indonesian Abstract

Luka didefinisikan sebagai rusaknya kesatuan jaringan pada tubuh. Perak memiliki banyak kegunaan dan memiliki potensi besar untuk pengobatan luka. Logam perak memiliki kemampuan merusak dinding sel bakteri, mengganggu metabolisme sel dan menghambat sintesis sel bakteri. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian air Ag (perak) terhadap kadar relatif TGF-β dan jumlah sel fibroblast sebagai penyembuhan luka insisi pada tikus putih (Rattus norvegicus). Penelitian ini bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL). Hewan model tikus (Rattus norvegicus) strain Wistar jantan dengan umur 8 – 10 minggu, berat badan 150 – 250 g, dibagi 4 kelompok. Kelompok negatif tanpa diinsisi, kelompok positif (terapi povidone iodine 0,5 mL), kelompok perlakuan 1 (terapi air Ag 0,5 mL) dan perlakuan 2 (terapi povidone iodine 0,5 mL dan air Ag 0,5 mL). Kadar relatif TGF-β dan perhitungan jumlah sel fibroblast diukur menggunakan metode flowcytometry kemudian dilakukan analisis dengan statistika ragam One-Way ANOVA, dan uji Tukey dengan tingkat kepercayaan 95%. Hasil penelitian menunjukkan bahwa pemberian terapi air Ag dapat meningkatkan secara signifikan kadar relatif TGF-β dan jumlah sel fibroblast. Kesimpulannya bahwa pemberian air Ag dapat meningkatkan kadar relatif TGF-β dan jumlah sel fibroblast untuk penyembuhan luka insisi pada tikus (Rattus novergicus).

English Abstract

Wounds are defined as damage to the unity of tissue in the body. Silver has many advantages and great potential for the treatment of wounds. Silver metal has the ability to damage bacterial cell walls, disrupt cell metabolism and inhibit bacterial cell synthesis. The aim of this study was to determine the effect of Ag (Silver) water on TGF-β expression and sel fibroblas in incision wound healing in rats (Rattus norvegicus) model animals. This research is experimental design that uses a completely randomized design (CRD). Animal models using rats (Rattus norvegicus) strain Wistar with age 8-10 weeks, weight 150-150 g, which were divided into 4 treatment groups. The negative group without incision, while the positive group (was treated with povidone iodine 0.5 mL). Treatment group 1 (was treated with Ag 0.5 mL) and treatment group 2 (was treated with povidone iodine 0.5mL and Ag water as much as 0.5 mL). TGF-β expression and calculation of fibroblast cells is measure by using the flowcytometry method and then analyzed with the statistics of the One-Way ANOVA variety, and the Tukey test with the confidence 95%. The results showed that of Ag water therapy could increase significant the relative levels of TGF-β and the number of fibroblast cells. It is concluded that Ag water could increase the relative levels of TGF-β and fibroblast cell count as healing incision wounds in rats (Rattus novergicus).
