Indonesian Abstract

Fase remodeling adalah salah satu parameter keberhasilan penyembuhan luka. Biochemical marker fase remodeling salah satunya adalah ALP. Gelatin ikan patin mengandung banyak kandungan asam amino salah satunya glisin dan glitamin yang menginduksi ALP pada osteoblastogenesis fase remodeling. Tujuan penelitian ini mengetahui pengaruh pemberian gelatin patin terhadap peningkatan ekspresi ALP pada penyembuhan luka pasca pencabutan gigi tikus putih.strain wistar. Penelitian menggunakan metode eksperimental Randomized Post Test Only Contol Group Design dilakukan pada hewan coba tikus putih. Sampel diambil secara random sampling kemudian dibagi menjadi 2 kelompok secara acak dengan 3 kontrol hari (hari ke-3, ke-5, dan ke-7). Kelompok kontrol (K) kelompok yang tidak diberi gelatin ikan patin dan kelompok perlakuan (P) kelompok yang diberi gelatin ikan patin. Pewarnaan yang digunakan adalah imunohistokimia. Pengamatan ekspresi ALP pada preparat HPA dengan mikroskop cahaya OLYMPUS pada 5 lapang pandang dengan perbesaran 400x. Uji ANOVA menunjukkan terdapat peningkatan jumlah ekspresi ALP yang signifikan pada kelompok perlakuan p<0.05, dengan puncak ekspresi ALP pada hari ke-7. Uji Post Hoc LSD membuktikan terdapat perbedaan bermakna antara kelompok kontrol dan perlakuan dengan p<0.05. Kesimpulannya adalah gelatin patin terbukti berpengaruh terhadap peningkatan jumlah ekspresi ALP pada penyembuhan luka pasca pencabutan gigi tikus putih strain wistar.

English Abstract

Remodeling phase is one of the success parameter of wound healing. One of the biochemical marker of remodeling phase is ALP. Gelatine of patine fish contains many of amino acids, one of them are arginine and glutamin, which can induce osteoblastogenesis process in wound healing. This study aims to prove that gelatine of patin fish can increase expression of ALP in wound healing after white rat’s tooth extraction. This study uses Experimental Randomized Post Test Only Control Group Design which is done to white rats. Random sampling methods is used to divided sample into 2 groups randomly with three control days (3rd day, 5th day, and 7th day). The control groups were not given the gelatine meanwhile the treatment group were given the gelatine. The number of ALP expression was calculated in 10 fields of view from HPA preparation with immunohistochemistry staining. Observation was conducted using microscope light OLYMPUS with 400x magnification. Anova test results that treatment group showed the significant increase with p<0.05, with the peak day of expressions in 7th day. Post Hoc LSD test results showed there was a significant difference between the control and treatment group with p< 0.05. The conclusion of this study is gelatine of patin (Pangasius djambal) has effect in increasing the expression of alkaline phosphatase (ALP) in wound healing after white rat’s (Rattus norvegicus) strain wistar tooth extraction.
