Indonesian Abstract

Amobilisasi enzim xilanase pada matriks bentonit dengan metode adsorpsi fisik bertujuan agar enzim mampu mempertahankan aktivitas dan dapat digunakan secara berulang maupun proses kontinyu. Xilanase diisolasi dari Aspergillus niger melalui proses fermentasi. Penelitian ini dilakukan untuk mengetahui kondisi optimum enzim xilanase amobil berdasarkan waktu pengocokan dan konsentrasi enzim. Variasi waktu pengocokan yang digunakan yaitu (1,2,3,4,5) jam dan variasi konsentrasi xilanase (0,148; 0,443; 0,738; 1,033; 1,328) mg/mL pada 0,1 g bentonin pada temperatur ruang. Enzim xilanase awal yang digunakan untuk amobilisasi memiliki kadar 1,475 mg/mL dengan aktivitas 22,19 unit. Hasil penelitian menunjukkan bahwa kondisi optimum amobilisasi xilanase dicapai pada saat waktu pengocokan 2 jam dan konsentrasi xilanase 0,443 mg/mL dengan jumlah xilanase teradsorpsi 1,574 mg dan aktivitas xilanase amobil sebesar 6,931 unit. Enzim xilanase amobil efisien digunakan hingga pemakaian ulang sebanyak 3 kali dengan aktivitas enzim sebesar 4,428 unit dan persentase efisiensi sebesar 64%. Berdasarkan analisis statistik menggunakan uji ANOVA satu arah, pengaruh waktu pengocokan dan konsentrasi enzim menunjukan perbedaan sangat nyata(0<0,01).

English Abstract

Xylanase enzyme immobilization in bentonite matrix with physical adsorption method so that the enzyme is able to maintain activity and can be used repeatedly and continuous processes. Xylanase was isolated from Aspergillus niger through a fermentation process. This study was conducted to determine the optimal conditions of xylanase enzymes based on the time of agitation and enzyme concentration. Variations in shaking time used were (1,2,3,4,5) hours and variations in xylanase concentration (0,148; 0,443; 0,738; 1,033; 1,328) mg / mL in 0,1 g bentonin at room temperature. The initial xylanase enzyme used for immobilization had a concentration of 1,475 mg / mL with an activity of 22,19 units. The results showed that the optimal conditions for xylanase immobilization were achieved at 2 hours shaking and xylanase concentration of 0,443 mg / mL with xylanase adsorbed 1,574 mg and immobilized xylanase activity at 6,931 units. Efficient immobilized xylanase enzyme was used until reuse 3 times with enzyme activity was 4,428 units and efficiency percentage was 64%. Based on statistical analysis using one-way ANOVA test, the effect of shaking time and enzyme concentration showed very significant differences(0 <0.01).
