Indonesian Abstract

Glomerulonefritis merupakan penyakit yang ditandai dengan adanya inflamasi dan proliferasi sel glomerulus ginjal. Streptokinase diproduksi oleh Streptococcus β hemolyticus yang berperan dalam patogenesis glomerulonefritis akut. Pemberian obat-obatan kimia seperti ACE Inhibitor pada kasus GNA dapat menyebabkan gangguan dan kerusakan pada ginjal sehingga penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol Blueberry (Vaccinium corrybosum) sebagai alternatif pengobatan pada kasus Glomerulonefritis Akut (GNA) terhadap kadar Malondialdehida (MDA) dan ekspresi Superoxida dismutase (SOD). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Tikus yang digunakan ialah tikus jantan strain wistar yang berumur 6-8 minggu dengan berat badan rata-rata 150-250 gram. Hewan coba dibagi dalam 5 kelompok perlakuan dengan 4 kali ulangan yaitu kontrol negatif (tikus sehat), kontrol positif (tikus diinduksi streptokinase 3 kali dosis 6000 IU), dan 3 kelompok perlakuan yang diinduksi streptokinase 3 kali dosis 6000 IU dan diterapi dengan ekstrak etanol Blueberry (dosis 500 mg/kg BB, 1000 mg/kg BB, 1500 mg/kg BB dengan pemberian satu kali sehari selama 14 hari). Setelah 37 hari, ginjal tikus diambil untuk pemeriksaan kadar MDA menggunakan spektrofotometer dan ekspresi SOD menggunakan flowcytometry. Analisis kuantitatif menggunakan One Way ANOVA dan dilanjutkan dengan uji lanjutan Tukey Test dengan taraf kepercayaan sebesar 95% atau α = 0.05. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol Blueberry dosis 500 mg/kg BB, 1000 mg/kg BB, dan 1500 mg/kg BB dapat digunakan sebagai terapi pada kasus glomerulonefritis akut hasil induksi streptokinase berdasarkan penurunan kadar Malondialdehida (MDA) dan peningkatan ekspresi Superoxide Dismutase (SOD) organ ginjal.

English Abstract

Glomerulonephritis is a disease characterized by inflammation and proliferation of renal glomerular kidney cells. Streptokinase is produced by Streptococcus β hemolyticus which plays a role in the pathogenesis of acute glomerulonephritis. Giving chemical drugs such as ACE inhibitors in the case of GNA can cause interference and damage to the kidneys so that this study aims to determine the effect of giving Blueberry ethanol extract (Vaccinium corrybosum) as an alternative treatment in cases of Acute Glomerulonephritis (GNA) to levels of Malondialdehyde (MDA) and expression of Superoxida dismutase (SOD). This study uses a Completely Randomized Design (CRD). The rats used were wistar male rats aged 6-8 weeks with an average body weight of 150-250 grams. The experimental animals were divided into 5 treatment groups with 4 replications namely negative controls (healthy rats), positive controls (rats induced streptokinase 3 times with the dose of 6000 IU), and 3 groups treated with streptokinase 3 times with the 6000 IU dose and treated with Blueberry ethanol extract (dosage of 500 mg/kg BB, 1000 mg/kg BB, 1500 mg/kg BB with one-day treatment for 14 days). After 37 days, rat kidneys were taken to examine the MDA levels using a spectrophotometer and SOD expression using flowcytometry. Quantitative analysis using One Way ANOVA and continued with the Tukey test follow-up test with a confidence level of 95% or α = 0.05. The results showed that treatment of Blueberry ethanol extract at a dose of 500 mg/kg BB, 1000 mg/kg BB, and 1500 mg/kg BB can be used as a therapy in cases of streptokinase induced acute glomerulonephritis based on decreased levels of Malondialdehyde (MDA) and increased expression of Superoxide Dismutase (SOD) kidney organs
