Indonesian Abstract

Shigella diperkirakan menyebabkan 80 hingga 165 juta kasus penyakit dan 600.000 kematian setiap tahun di seluruh dunia. Di Indonesia, 29% kematian yang disebabkan oleh diare pada kelompok usia 1 hingga 4 tahun adalah disentri basiler. Shigellosis adalah infeksi yang bekerja pada saluran pencernaan yang disebabkan oleh Shigella spp. Ketahanan spesies Shigella terhadap beberapa antibiotik telah dilaporkan di seluruh dunia. Oleh karena itu, mengembangkan pengobatan alternatif dari tanaman obat sangat penting. Salah satu tanaman obat alami yang memiliki potensi antibakteri adalah daun delima (Punica granatum). Daun delima mengandung konstituen aktif seperti tanin, flavonoid dan saponin yang disebut memiliki efek antimikrobial. Tujuan dari penelitian ini adalah untuk membuktikan efek antimikroba ekstrak etanol daun delima terhadap pertumbuhan Shigella dysentriae sehingga nantinya dapat digunakan sebagai pengobatan shigellosis. Metode yang digunakan untuk penelitian ini adalah metode dilusi agar dengan empat kali pengulangan menggunakan berbagai konsentrasi ekstrak etanol daun delima yaitu 0% v/v, 0,5% v/v, 1% v/v, 2% v/v, 3%v /v dan 4% v/v terhadap Shigella dysentriae. Hasilnya menunjukkan bahwa tidak ada pertumbuhan yang diamati pada 3% v/v dan 4% v/v. Hasil penelitian menunjukkan bahwa ada perbedaan derajat pertumbuhan bakteri antara berbagai konsentrasi ekstrak (Kruskal-Wallis, P=0,000) dan ada hubungan antara berbagai konsentrasi ekstrak dengan derajat pertumbuhan bakteri (korelasi Spearman, R = -0,950, P = 0,000). KHM ekstrak ethanol daun delima sebagai agen antimikroba terhadap Shigella dysentriae ada pada konsentrasi 3%v/v. Berdasarkan hasil penelitian, dapat disimpulkan bahwa ekstrak etanol daun delima memiliki efek antimikroba terhadap pertumbuhan Shigella dysentriae secara in vitro dengan KHM 3%v/v..

English Abstract

Shigella is estimate to cause 80 to 165 million cases of disease and 600,000 deaths annually worldwide. In Indonesia, 29% of death caused by diarrhea in the age group of 1 to 4 years old were bacillary dysentery. Shigellosis is an infection acting on the digestive tract caused by Shigella spp. Resistance of Shigella species to some antibiotics has been reported worldwide. Hence, develop alternative treatment from medicinal plants is essential. One of the natural medicinal plants that has antibacterial potential is pomegranate leaf (Punica granatum). Pomegranate leaf contains active constituents such as tannin, flavonoid and saponin that have antimicrobial effect. The objective of the research is to prove the antimicrobial effect of pomegranate leaf ethanol extract against the growth of Shigella dysentriae in hope that it can be used as shigellosis treatment in the future. Method used for the research is agar dilution method with four times replication using various of concentration of pomegranate leaf ethanol extract which is 0%v/v, 0.5%v/v, 1%v/v, 2%v/v, 3%v/v and 4%v/v against Shigella dysentriae. The results indicate that there is no growth observed on 3%v/v and 4%v/v. Research result shows that there is difference in bacteria growth degree between different extract concentration (Kruskal-Wallis, P=0.000) and there is relationship between bacteria growth degree with different extract concentration (Spearman correlation, R=-0.950,P=0.000). MIC of pomegranate leaf ethanol extract as antimicrobial agent against Shigella dysentriae is observed at 3%v/v. Based on the results, can be concluded that pomegranate leaf ethanol extract indeed has antimicrobial effect against the growth of Shigella dysentriae in vitro with 3%v/v MIC.
