Indonesian Abstract

Ekstrak kulit manggis kaya akan senyawa xanthone dan dapat digunakan sebagai alternatif terapi dermatitis atopik. Dengan memanfaatkan sistem penghantaran obat berbasis vesikel lipid transfersom dapat ditingkatkan penghantaran dan efek terapetiknya. Transfersom tersusun dari fosfolipid dan edge-activator berupa surfaktan rantai tunggal. Surfaktan dapat mempengaruhi sifat fisikokimia vesikel, sehingga menjadi titik kritis dalam formulasi transfersom. Penelitian ini dilakukan untuk mengetahui pengaruh variasi surfaktan terhadap ukuran partikel transfersom ekstrak kulit buah manggis. Transfersom ekstrak kulit buah manggis dipreparasi menggunakan 3 variasi surfaktan dengan HLB berbeda-beda yaitu Span 80 (HLB 4,3), kombinasi Span 80 dan Tween 80 (HLBcampur 10), dan Tween 80 (HLB 15), dengan metode hidrasi lapis tipis. Transfersom dievaluasi organoleptis, pH, ukuran dan distribusi ukuran, dan potensial zeta. Formula optimum dievaluasi stabilitas lebih lanjut. Data hasil penelitian dianalisis menggunakan analisa ragam satu arah (One-way ANOVA) dilanjutkan uji Post-hoc Tukey HSD, uji beda dua sampel berpasangan, dan uji korelasi pearson. Semua formula menghasilkan pH dalam rentang 6,36-7,13, ukuran vesikel <450 nm, indeks polidispersitas <0,3, dan potensial zeta <-60 mV. Formulasi dengan tween 80 mempunyai ukuran paling kecil dan stabilitas yang baik pada pH, namun tidak stabil dalam ukuran, distribusi ukuran dan potensial zeta. Analisis statistik menunjukkan variabel bebas mempunyai pengaruh signifikan terhadap variabel terikat (p<0,05). Berdasarkan hasil penelitian, disimpulkan bahwa semakin tinggi nilai HLB maka semakin kecil ukuran vesikel yang dihasilkan.

English Abstract

Mangosteen pericarp extract is rich of xanthones and able to be an alternative treatment of atopic dermatitis. The use of lipid vesicles-based drug delivery system helps to improve its delivery and therapeutic effect. Transfersomes is formed by combination of phospholipids and edge-activators such as single chain surfactants. Surfactant affects its physicochemical characteristic, therefore become a critical point in transfersomes formulation. This study was aimed to investigate the effect of surfactant variation on vesicle size of mangosteen pericarp extract transfersomes. The transfersomes were prepared by thin film hydration method using various surfactant with different HLB such as Span 80 (HLB 4,3), Span 80 and Tween 80 combination (HLBmixed 10), Tween 80 (HLB 15), and evaluated for organoleptic, pH, vesicle size and size distribution, and zeta potential. The selected optimum formulation was further evaluated for stability studies. The data was analyzed by One-way ANOVA followed by Tukey HSD Post-hoc Test, Paired T-Test, and Pearson Correlation Test. All formulations gave pH value ranging from 6,36 to 7,13, vesicle size <450 nm, polydispersity index <0,3, and zeta potential <-60 mV. Formulation with tween 80 had smallest vesicle size and good stability on pH, but unstable on vesicle size, size distribution and zeta potential. Statistical analysis showed independent variable had a significant effect on the dependent variable (p<0,05). It is concluded that the higher HLB value, the smaller vesicle size.
