Indonesian Abstract

Di dalam asap rokok terdapat banyak radikal bebas yang berbahaya bagi ibu hamil. Paparan asap rokok selama kehamilan mengakibatkan stres oksidatif dan peroksidasi lipid plasenta dengan hasil akhir Malondialdehyde (MDA). Terjadinya peroksidasi lipid plasenta menyebabkan kerusakan sel endotel plasenta. Brokoli mengandung senyawa antioksidan yang dapat menangkap radikal bebas sehinggamencegah terjadinya stres oksidatif. Tujuan penelitian ini untuk mengetahui efek ekstrak etanol brokoli pada kadar MDA plasenta tikus bunting yang terpapar asap rokok. Rancangan penelitian yang digunakan adalah Randomized Post Test Only Control Group Design dengan menggunakan tikus bunting. Sampel terbagi dalam 2 kelompok kontrol (K (-)=tanpa perlakuan, K (+)=dipapar asap rokok) dan 3 kelompok perlakuan dengan paparan asap rokok serta ekstrak etanol brokoli dengan dosis berbeda (P1=100, P2=200, P3=400 mg/kgBB/hari). Pemberian ekstrak etanol brokoli dimulai hari ke 1-19 kebuntingan sedangkan paparan asap rokok dimulai hari ke 7-19 kebuntingan. Hasil uji One Way ANOVA didapatkan adanya perbedaan antar kelompok (p=0,000). Pada analisa Tukey HSD, hasil perbandingan K (+) dengan P1, P2, dan P3 diperoleh p = 0,000 yang berarti ada perbedaan secarasbermakna. Uji korelasi Pearson menunjukkan arah hubungan negatif yang berari semakin besar ekstrak etanol brokoli yang diberikan, semakin rendah kadar MDA plasenta. Ekstrak etanol brokoli dapat menurunkan kadar MDA plasenta tikus bunting yang terpapar asap rokok dan dosis efektif dalam penelitian ini adalah 400 mg/kgBB.

English Abstract

In cigarette smoke there are many free radicals that are harmful to pregnant women. Exposure to cigarette smoke during pregnancy results in oxidative stress and placental lipid peroxidation with the end result of Malondialdehyde (MDA). The occurrence of placental lipid peroxidation can cause placental endothelial cell damage. Broccoli contains antioxidant compounds that can capture free radicals so prevent oxidative stress. This study was conducted to determine effect of broccoli ethanol extract on MDA levels in placental pregnant mice exposed to cigarette smoke. The research design used was Randomized Post Test Only Control Group Design using pregnant mice. The sample was divided into 2 control groups (K (-) = no treatment, K (+) = exposed to cigarette smoke) and 3 treatment groups with exposure to cigarette smoke and broccoli ethanol extract with different doses (P1 = 100, P2 = 200, P3 = 400 mg / kgBW / day). The administration of broccoli ethanol extract starts from 1-19 days, while exposure to cigarette smoke begins on the 7-19 day of pregnancy. The One Way ANOVA test results found differences between groups (p = 0,000). In the Tukey HSD analysis, the results of the K (+) comparison with P1, P2, and P3 obtained p = 0,000 which means there are significant differences. The Pearson correlation test shows direction of the negative relationship which means the greater broccoli ethanol extract given, the lower placental MDA level. Broccoli ethanol extract can reduce MDA level of placenta in pregnant mice exposed to cigarette smoke and effective dose in this study is 400 mg / kgBW.
