Indonesian Abstract

Penelitian ini bertujuan untuk mengetahui proses metilasi pada sequence protein EEF1A1 dan gen EEF1A1 serta mengetahui pengaruh reaksi metilasi yang terjadi pada protein EEF1A1 dan gen EEF1A1 dengan menggunakan katalis METTL10 secara in-silico. Proses analisis in-silico dengan metode molecular docking dilakukan dalam beberapa tahapan, yaitu 1) Penentuan protein EEF1A1, gen EEF1A1, METTL10, struktur model AdoMet, dan struktur model Fe4S4, 2) Preparasi FASTA protein EEF1A1 dan METTL10, 3) Perlakuan metode docking pada protein EEF1A1 dan gen EEF1A1, dan 4) Analisis molecular docking: energi bebas Gibbs dan konstanta inhibisi. Hasil yang diperoleh dari penelitian ini berupa nilai energi bebas Gibbs (ΔG). Semakin kecil nilai ΔG maka ikatan antara reseptor dengan ligan akan semakin kuat dan stabil. Selain itu, proses metilasi juga semakin baik jika nilai ΔG kecil (ΔG < 0). Nilai ΔG yang didapatkan dari reaksi metilasi pada protein EEF1A1 menggunakan katalis METTL10 sebesar -0,08 kcal/mol, sedangkan nilai ΔG yang didapatkan dari reaksi metilasi pada gen EEF1A1 menggunakan katalis METTL10 sebesar -0,82 kcal/mol. Reaksi metilasi lebih cenderung pada gen EEF1A1, karena nilai ΔG pada gen EEF1A1 khususnya pada basa nitrogen timin lebih kecil jika dibandingkan dengan nilai ΔG pada protein EEF1A1. Metilasi pada gen EEF1A1 menyebabkan terjadinya mutasi pada gen EEF1A1, sehingga protein EEF1A1 yang berperan dalam proses apoptosis menjadi terganggu, maka sel akan cenderung mengalami proliferasi terus-menerus. Oleh karena itu proses metilasi pada gen EEF1A1 yang dikatalis oleh METTL10 dapat menguatkan bahwa gen EEF1A1 dan protein EEF1A1 terkait dengan pertumbuhan sel kanker.

English Abstract

This research was conducted with purpose to determine the process of methylation in EEF1A1 protein sequence and EEF1A1 gene and to know the effect of methylation reaction occurring on EEF1A1 protein and EEF1A1 gene by using METTL10 catalyst in-silico. The process of in-silico analysis with molecular docking method is done in several steps 1) Determination of EEF1A1 protein, EEF1A1 gene, METTL10, AdoMet model structure, and Fe4S4 model structure, 2) Preparation FASTA of EEF1A1 and METTL10 proteins, 3) Treatment of docking method on EEF1A1 protein and EEF1A1 gene, and 4) Molecular docking analysis: Gibbs free energy and inhibition constant. The result of this research is Gibbs free energy (ΔG). The smaller the value of ΔG the bond between the receptor and the ligand will be stronger and more stable. In addition, the methylation process is also better if the value of ΔG is small (ΔG < 0). The ΔG values obtained from the methylation reaction of the EEF1A1 protein used METTL10 catalysts of -0.08 kcal/mol, while the ΔG values obtained from the methylation reaction in the EEF1A1 gene used METTL10 catalysts of -0.82 kcal/mol. The methylation reaction is more likely in the EEF1A1 gene, since the value of ΔG in the EEF1A1 gene is smaller when compared to the ΔG value of EEF1A1 protein. The methylation reaction is more likely in the EEF1A1 gene, since the value of ΔG in the EEF1A1 gene especially on the thymine nitrogen base is less than the ΔG value of EEF1A1 protein. Methylation of the EEF1A1 gene causes mutations in the EEF1A1 gene, so EEF1A1 proteins that plays a role in apoptosis process becomes disrupted, then the cells will tend to experience continuous proliferation. Therefore, the methylation process in EEF1A1 gene catalyzed by METTL10 can confirm that EEF1A1 protein and EEF1A1 gene are associated with the growth of cancer cells.
