Indonesian Abstract

Penelitian ini bertujuan untuk menganalisis profil peptida kolagen sisik ikan gabus (Channa striata) dari hasil ekstraksi enzimatis menggunakan enzim papain. Metode penelitian ini diawali isolasi enzim papain dari getah pepaya (Carica papaya L.). Crude papain dipresipitasi menggunakan etanol absolut dan crude papain non-presipitasi digunakan untuk ekstraksi enzimatis sisik ikan gabus setelah pre-treatment oleh NaOH dan Na2EDTA. Hasil ekstraksi dibedakan menjadi dua kelompok sampel, yaitu hidrolisat kolagen non-freeze dry dan freeze dry. Konsentrasi hidrolisat kolagen dan crude papain ditentukan melalui uji Bradford. Penentuan berat molekul peptida berdasarkan SDS-PAGE konsentrasi separating gel 15% dengan kontrol crude papain. Kurva standar protein dan berat molekul (BM) ditampilkan dalam bentuk grafik dengan Ms. Excel. Perbedaan rerata konsentrasi sampel ditentukan melalui IndependentSamples T Test dengan selang kepercayaan 95%. Hasil menunjukkan bahwa terdapat perbedaan rerata konsentrasi antara crude papain non-presipitasi dengan presipitasi, sedangkan rerata konsentrasi hidrolisat kolagen non-freeze dry dan freeze dry tidak berbeda signifikan berdasarkan analisis statistik dengan Independent-Sample T Test (p<0,05). Hasil visualisasi SDS-PAGE pada sampel hidrolisat kolagen hanya tampak band dengan BM 24 kDa yang teridentifikasi sebagai papain. Profil peptida kolagen dalam hidrolisat kolagen sisik ikan gabus hasil ekstraksi menggunakan enzim papain memiliki berat molekul <10 kDa pada penelitian ini.

English Abstract

The aims of this experiment to analyze the peptide profile of snakehead fish scales (Channa striata) derived from enzymatic extraction by papain enzyme. The method begins with the isolation of papain enzyme from papaya latex (Carica papaya L.). Crude papain was precipitated using absolute ethanol and non-precipitation crude papain was used for the enzymatic extraction of fish scales after pre-treatment by NaOH and Na2EDTA. The extraction results were divided into two groups of samples, collagen hydrolysate nonfreeze dry and freeze dry. The concentration of collagen hydrolysate and crude papain was determined by Bradford assay. Peptide molecular weight was estimated based on SDS-PAGE with 15% separating gel concentration using crude papain as control. Standard protein curve and molecular weight (MW) are shown in graphical form with Ms. Excel. The average difference of sample concentration was determined through Independent-Samples T Test with 95% confidence interval. The results showed that there was a difference of mean concentration between non-precipitation crude papain with precipitation, whereas the mean of non-freeze dry and freeze dry collagen hydrolysate concentration was not significantly different based on statistical analysis with Independent-Sample T Test (p <0,05). The SDS-PAGE visualization results in collagen hydrolysate samples only appeared with a band of 24 kDa identified as papain. Profile of collagen peptide in hydrolysate collagen of snakehead fish scales extracted by papain enzyme have molecular weight <10 kDa in this study
