Indonesian Abstract

MethicillinQResistant Staphylococcus aureus (MRSA)ImerupakanQsalah satu patogen penyebabQinfeksi nosokomial denganQkasus resistensi antibiotika yang tinggi. ResistensiQbakteri tersebut diakibatkan prosesQpembentukan β- laktamase oleh bakteri StaphylococcusQaureus. Biji hijau kopi robusta (Coffea canephora) memilikiQsenyawa seperti asam klorogenat, kafein dan senyawa fenol, yang dapat berfungsi sebagai antimikrobaQpada bakteri Methicillin Resistant Staphylococcus aureus. Tujuan dari penelitianQini adalah untuk mengetahui efek dari pemberianQekstrak padaQbiji tumbuhan kopi robusta (Coffea canephora) terhadap bakteri Methicillin Resistant Staphylococcus aureus (MRSA) secara in vitro. Penelitian ini menggunakan metode difusiQsumuran dengan empat kali pengulangan. Terdapat 6 kelompok dengan 5 kelompok perlakuan dengan konsentrasi 20%; 40%; 60%; 80%; 100%, serta kontrolQnegatif berisi ekstrak konsentrasi 0%. Hasil dari perlakuanQtersebut didokumentasi lalu menghitung zona hambat menggunakanQjangka sorong denganQketelitian 1mm. Hasil penelitianQmenunjukkan bahwa, terdapat peningkatan diameter zona hambat yang berbanding lurus dengan konsentrasi ekstrak. Uji One WayQAnova didapatkan signifikansi p=0,000 (p<0,05) disertai hasil korelasi yang kuat antara konsentrasi danQzona hambat value (korelasi, p<0,05). Pada hasil akhir dengan uji regresi didapatkan R Square (R2) sebesar 0.691. KesimpulanQpenelitian ini adalah ekstrak etanol biji hijau kopi robusta dapatQmenghambat pertumbuhan bakteri MethicillinQResistant Staphylococcus secara in vitro denganQsignifikan.

English Abstract

MethicillinqResistant Staphylococcus aureus (MRSA) is oneqof the pathogens that cause nosocomial infections with casesqof high antibiotic resistance. The bacterial resistance is caused by the formation of β-lactamase by the Staphylococcus aureus bacteria. Greenqbeans of robusta coffee (Coffea canephora) haveqcompounds such as chlorogenic acid, caffeine and phenol compounds, which can function as antimicrobialsqon the Methicillin Resistant Staphylococcus aureus bacteria. The purposeqof this study was to determine theq effect of giving extracts on the seeds of Robustaqcoffee (Coffea canephora) to theq bacteria Methicillin Resistant Staphylococcus aureus (MRSA) in vitro. This studyquses a well diffusion method with four repetitions. There are 6 groups with 5 treatmentqgroups with aqconcentration of 20%; 40%; 60%; 80%; 100%, and negative controls contain extracts of 0% concentration. The resultsqof the treatment are documented and then calculateqthe inhibition zone using a calipers with accuracy of 1mm. The results showed that thereqwas an increase in the diameter of the inhibition zone which was directlyqproportional to the concentration of extract. One Way Anova test obtainedqsignificance p = 0,000 (p <0.05) accompanied by a strong correlation betweenqconcentration and inhibition value zone (correlation, p <0.05). In the finalqresult, the regression testqshows that R Square (R2) is 0.691. The conclusion ofqthis study is that the ethanol extract of robusta coffee green seeds can significantly inhibit the growth of Methicillin Resistant Staphylococcus bacteria in vitro.
