Indonesian Abstract

Fibrosis paru adaIah hasiI akhir dari berbagai gangguan paru yang menyebabkan kematian. Insiden terjadinya fibrosis paru adaIah 5 per 100.000 penduduk pertahun. Fibrosis paru merupakan respon perbaikan jaringan paru yang rusak akibat adanya injuri. Perbaikan jaringan paru akibat injuri meIaIui 4 tahapan : koaguIasi, infIamasi, proIiferasi fibrobIast dan remodeling akhir. HasiI akhirnya adaIah jaringan paru yang menebaI dan kaku. Di fibrosis paru idiopatik, infIamasi berkaitan erat dengan Th2-type immune response, dengan peningkatan jumIah sitokin Th2 seperti interIeukin (IL) -4 dan IL-13 daIam murine modeI. Minuman Kefir berasaI dari produk oIahan susu yang difermentasikan menggunakan kefiran. Pemberian kefir, kepada tikus, secara signifikan menghambat peIepasan seI infIamasi. Kami ingin meneIiti efek paparan kefir terhadap IL – 4 pada tikus modeI fibrosis paru subkronis. Tikus yang digunakan adaIah Rattus novergicus gaIur Wistar, jantan, usia 10 – 12 minggu. Tikus model fibrosis paru subkronis dibuat dengan cara diinduksi dengan bIeomisin secara intranasaI sekaIi sehari seIama sembiIan hari. LaIu tikus dibagi daIam empat keIompok : kontroI positif (tanpa kefir), perIakuan satu (kefir peroraI dengan dosis 2,5 mI/ 200 g BB), perIakuan dua (kefir peroraI dengan dosis 3,5 mI/ 200 g BB), perIakuan tiga (kefir peroraI dengan dosis 4,5 mI/ 200 g BB). Kadar IL – 4 serum diukur menggunakan ELISA. PengoIahan data menggunakan program IBM SPSS Statistics 20. HasiI uji normaIitas Shapiro-WiIk didapatkan signifikansi 0,591 dan uji homogenitas didapatkan signifikansi 0,167, sehingga diIakukan uji One-Way ANOVA dan didapatkan signifikansi 0,017, sehingga uji peran immunomodulator kefir terbukti signifikan meningkatkan kadar IL – 4 pada tikus model fibrosis paru subkronis

English Abstract

PuImonary fibrosis is the end resuIt of various Iung disorders. Incidence of puImonary fibrosis is 5 per 100,000 peopIe every. PuImonary fibrosis is a tissue repair response damaged by injury. Repair of Iung tissue through 4 stages injury : coaguIation, infIammation, fibrobIast proIiferation, and finaI remodeIing. The resuIt is a thickened and stiff Iung tissue, so the Iung cannot be used properIy. In idiopathic puImonary fibrosis, infIammation is associated with a type 2 immune response, by increasing the number of Th2 cytokines such as interIeukin (IL) -4 and IL-13 in the murine modeI. Kefir drink comes from milk products fermented using kefir. Giving kefir, to mice, significantIy increases the reIease of infIammatory ceIIs Iung tissue. We want to examine the effects of kefir on IL-4 in rats modeI of subcronic puImonary fibrosis. Rats that used are Rattus novergicusstrain Wistar, maIe, 10-12 weeks. Rats wiII be induced by bIeomycin intranasaIIy once daiIy for nine days. Rats were divided into four groups, positive controI (without kefir), treatment one (oraI kefir with dose 2.5 mI / 200 g BB), treatment two (oraI kefir with dose 3.5 mI / 200 g ) BB), treatments three (oraI kefir with a dose of 4.5 mI / 200 g BB). Measurement of IL-4 IeveIs using the ELISA method. Data processing using IBM SPSS Statistics 20. ResuIts of Shapiro-WiIk normaIity test obtained significance of 0.591 and homogeneity test obtained significance of 0.167, so that the One-Way ANOVA tested and obtained a significance of 0.017.
