Indonesian Abstract

Diet aterogenik memiliki risiko meningkatkan beberapa faktor metabolit tubuh, seperti menyebabkan resisten insulin di jaringan perifer maupun ketidakseimbangan profil lipid yang berakibat pada ketidaknormalan sintesis trigliserida. Bila trigliserida terakumulasi pada organ hepar, maka dapat menyebabkan terjadinya perlemakan hati, salah satunya non alcoholic fatty liver disease (NAFLD). Derajat keparahan NAFLD berkaitan dengan peningkatan kadar mediator inflamasi, salah satunya TNF-α. Alga (Sargassum sp.) mengandung beragam zat bioaktif, seperti fucoidan, flavonoid, triterpenoid, tannin, phlorotannin, dan polifenol, di mana dalam kandungannya ada yang mampu bersifat sebagai antioksidan, anti-inflamasi, anti-bakteri, dan anti-tumor. Penelitian ini menggunakan metode randomized post test only controlled group design yang bertujuan untuk mengetahui pengaruh pemberian ekstrak alga coklat (Sargassum sp.) terhadap ekpresi kadar TNF-α hepar pada tikus wistar dengan diet aterogenik. Sampel yang digunakan berjumlah 30 ekor tikus wistar berjenis kelamin jantan dan dibagi menjadi enam kelompok, yaitu Kelompok Negatif (diet normal), Kelompok Positif (diet aterogenik), Kelompok Dosis Perlakuan 1, Dosis Perlakuan 2, dan Dosis Perlakuan 3 yang diinduksi diet aterogenik dan ekstrak alga coklat (Sargassum sp.) dosis 150mg/kgBB, 300mg/kgBB, dan 600mg/kgBB, serta Kelompok Simvastatin yang diberi simvastatin dosis 4mg/kgBB mulai dari minggu ke-5 penelitian. Penelitian dilakukan selama 12 minggu dengan parameter mengacu pada ekpresi TNF-α. Pengukuran ekspresi TNF-α menggunakan imunohistokimia (IHK). Hasil uji statistik One-Way Anova didapatkan nilaip=0,000 (p < 0,05) yang berarti terdapat perbedaan bermakna antar kelompok perlakuan. Dari hasil penelitian ini, dapat disimpulkan bahwa ekstrak alga coklat (Sargassum sp.) mampu menurunkan ekpresi TNF-α hepar pada tikus wistar dengan diet aterogenik

English Abstract

Atherogenic diets increase the risk of several metabolic disorders such as insulin resistance in peripheral tissues and imbalance of lipid profiles that cause abnormal triglyceride synthesis. As triglycerides accumulates in the liver, it can causes fatty liver, as seen in non-alcoholic fatty liver disease (NAFLD). The severity of NAFLD is associated with an elevation of inflammatory mediator level, including TNF-α. Sargassum sp. contains various bioactive substances, such as fucoidan, flavonoids, triterpenoids, tannins, phlorotannins, and polyphenols, which act as antioxidants, anti-inflammatory, anti-bacterial, and anti-tumor. This study was designed as a randomized post-test only controlled group aimed to investigate the effect of brown algae extract (Sargassum sp.) administration toward hepatic TNF-α expression in atherogenic diet Wistar rats. As many as 30 male Wistar rats were divided into six groups as follows: negative control (normal diet), positive control (atherogenic diet), treatment 1, 2, and 3 (atherogenic diet + brown algae extract (Sargassum sp.) dosage 150 mg/kgBB, 300 mg/kgBB, and 600 mg/kgBB, respectively), and simvastatin (atherogenic diet + simvastatin dosage 4 mg/kgBB). Atherogenic diet was administered for 12 weeks and all treatments were started from the 5th week of the study. Measurement of TNF-α expression using immunohistochemistry (CPI). The results showed that there were treatment of Sargassum sp. extract attenuates the elevation of TNF- α expression caused by atherogenic diet (p < 0.05) and this finding was not significantly different with simvastatin-treated group (p > 0.05). We concluded that brown algae extract (Sargassum sp.) potential for reducing hepatic TNF-α expression in atherogenic diet Wistar rats.
