Indonesian Abstract

Kafein (1,3,7-trimetilxantin) merupakan salah satu zat yang paling banyak dikonsumsi dan tersebar dalam berbagai macam makanan dan minuman, termasuk beberapa jenis obat-obatan.Kafein dapat melewati sawar plasenta menuju cairan amnion dan janin.Kafein dapat menimbulkan keadaan stress oksidatif dan secara lebih lanjut keadaan stress oksidatif selama ini dipercaya dapat meningkatkan risiko terjadinya penyakit kardiovaskuler. Disisi lain, saat ini terdapat buah yang cukup populer dengan kandungan senyawa bioaktif antioksidan, yakni buah naga merah (Hylocereus polyrhizus). Tujuan penelitian ini adalah untuk membuktikan efek kafein terhadap denyut jantung janin dan pengaruh pemberian ekstrak buah naga merah dalam menghambat peningkatan denyut jantung akibat kafein menggunakan embrio ikan zebra sebagai model sampel. Embrio ikan zebra dibagi dalam enam kelompok berbeda berdasarkan paparannya, kontrol (tanpa paparan kafein dan esktrak buah naga merah), kafein 20 mg/L (kontrol negatif), kafein 20 mg/L kombinasi vitamin C 44 mg/L (kontrol positif), kafein 20 mg/L kombinasi esktrak buah naga merah dosis 29 mg/L, 58 mg/L, dan 87 mg/L (perlakuan 1, 2, 3). Uji Independen T-tes menunjukkan perbedaan signifikan rata-rata frekuensi denyut jantung embrio ikan zebra antara kelompok kontrol dan kelompok paparan kafein (p = 0.012), sedangkan uji One Way ANOVA didapatkan perbedaan tidak signifikan rata-rata frekuensi denyut jantung embrio ikan zebra antara kelompok paparan kafein dengan paparan kafein kombinasi ekstrak buah naga merah (p = 0.246). Hal ini menunjukkan bahwa kafein bekerja dalam berbagai macam reseptor, sehingga pemberian antioksidan tidak cukup mampu dalam menghambar peningkatan denyut jantung embrio ikan zebra akibat paparan kafein.

English Abstract

Caffeine (1,3,7-trymethylxanthine) is a substance which is widely consumed and it contains on many kind of foods and beverages also several medicinal products. Caffeine can pass placenta barrier freely. Caffeine consumed can stimulate oxidative stress. Oxidative stress was believed could increase the risk of cardiovascular diseases. In the other hand, red dragon fruit (Hylocereus polyrhizus) is recently popular due its bioactive antioxidant substances against the oxidative stress. The aim of this study is to examine the effect of caffeine to fetal heart rate and the effect of red dragon fruit extract to caffeine exposure on fetal heart rate using zebrafish embryos as animal model. Fertilized zebrafish eggs were divided into six groups regard on its exposure, control (without caffeine and red dragon fruit extract), caffeine 20 mg/L (negative control), caffeine 20 mg/L with vitamin C 44 mg/L (positive control), and caffeine 20 mg/L combines with red dragon fruit extract dose 29 mg/L, 58 mg/L, 87 mg/L (treatment 1, 2, 3). Based on statistical analyze using Inependent T-test there is a significant difference of zebrafish embryos’ heart rate between control and caffeine exposure group (p = 0.012), meanwhile the One Way ANOVA analyze shows there is no significantlly difference of zebrafish embryos’ heart rate among caffeine exposure group and three of caffeine combines with red dragon fruit extract groups (p = 0.246). We noted that caffeine acts on many kind receptors in the body and that is why antioxidants are impossible to prevent the increased heart rate due to caffeine exposure.
