Abstract
Hidroklorotiazid (HCT) merupakan obat anti hipertensi golongan thiazide yaitu diuretic yang memiliki potensi menurunkan tekanan darah dengan menurunkan jumlah cairan dalam tubuh. HCT memiliki efek samping stomach cramps dan memiliki waktu paruh (t1/2) hanya 5,6 sampai 14,8 jam. Hal ini akan mempengaruhi pemberian secara oral hidroklorotiazid kepada pasien. Efek samping yang terjadi dan frekuensi pemberian obat yang banyak kepada pasien dapat menurunkan kepatuhan penggunaan obat tersebut. Penelitian ini bertujuan untuk menegambangkan formula nanoemulsi HCT dalam sediaan patch. Nanoemulsi HCT didapatkan dengan metode Self-Nanoemulsifying Drug Delivery System (SNEDDS). Nanoemulsi HCT kemudian diformulasikan kedalam sediaan patch menggunakan metode solvent evaporation menggunakan variasi perbandingan HPMC dan EC yaitu 3 : 1; 1 : 1; dan 3 : 1. Patch dievaluasi organoleptis, keseragaman bobot, ketebalan, daya lipat, % drug content dan In vitro permeation. Formula patch yang optimum adalah formula dengan perbandingan polimer HPMC : EC 1 : 1 yang berwarna transparant,tidak berbau, halus dan flexible, dengan bobot 1,2877 ± 0,6099 gram dan tebal 0,5973 ± 0,021 mm, memiliki ketahanan daya lipat > 200 kali dan % drug content yaitu 90.081 ± 1,607. Uji In vitro permeation menunjukkan % terdifusi puncak terdapat pada jam ke-8 sebesar 5,076% dan % cumulative drug release sebesar 34,5771 ± 2,6988 % selama 12 jam. Analisis kinetik menunjukkan pelepasan obat mengikuti model Higuchi.
English Abstract
Hydrochlorothiazide (HCT) is a anti-hypertention drug that belongs to thiazide class. HCT is a diuretic that potential reduce a blood pressure by decrease the amount of body fluids. HCT has side effect causing stomach cramps dan half time (t1/2) 5,6 until 14,8 hours. That can effected oral administration of HCT to patients. Side effect and increasing frequency of drug administration can reduce patients’ compliance. The aim of this research is to develop HCT nanoemulsion formula in patch preparation. HCT nanoemulsion was made using Self-Nanoemylsifying Drug Delivery System (SNEEDS) method.HCT nanoemulsion was formulated into patch using solvent evaporation method with variation of concentration ratio HPMC : EC, that was 3 : 1; 1 : 1; and 1 : 3. The optimum patch formula was formula with ratio HPMC and EC polymer is 1 : 1 . The organoleptic of patch was transparant colour, odorless, dan flexible, folding endurance > 200x. and % drug content was 90.081 ± 1,607 %. In vitro permeation showed that % drug release had a peak in 8th hours with % release 5,076% % cumulative drug release 34,5771 ± 2,6988 % for 12 hours. Kinetic analysis showed that drug release of optimum formula was adequately described by Higuchi model.
