Indonesian Abstract

Penggunaan doksorubisin sebagai kemoterapi kanker payudara sering kali dibatasi akibat toksisitasnya terhadap jantung. Rumput lidah ular (Hedyotis diffusa Willd.) merupakan obat bahan alam yang banyak diresepkan dalam herbal Cina untuk pengobatan kanker payudara. Hedyotis diffusa Willd. dan doksorubisin dikombinasikan sehingga dapat menurunkan dosis doksorubisin yang digunakan. Penelitian ini dilakukan untuk mengetahui nilai IC50 Hedyotis diffusa Willd. dan doksorubisin, efek kombinasi yang ditimbulkan, serta kombinasi konsentrasi yang dapat menimbulkan efek sinergis. Hedyotis diffusa Willd. diekstraksi dengan metode maserasi dalam pelarut etanol 80%. Kultur sel MCF-7 diberikan perlakuan dengan 7 seri konsentrasi selama 24 jam, kemudian nilai IC50 diukur menggunakan metode MTT. Kombinasi diberikan dengan konsentrasi 1/2 IC50, 3/8 IC50, 1/4 IC50, dan 1/8 IC50 kemudian dihitung nilai CI. Nilai IC50 ekstrak Hedyotis diffusa Willd. sebesar 930 ± 30,143 μg/ml (p<0,0001) dan doksorubisin sebesar 490 ± 83,293 nM (p<0,0001). Kombinasi ekstrak Hedyotis diffusa Willd. dan doksorubisin didapatkan efek sinergis pada kombinasi konsentrasi doksorubisin 61 nM dengan ekstrak Hedyotis diffusa Willd. 116 μg/ml dan 232 μg/ml. Berdasarkan hasil tersebut, diketahui bahwa kombinasi ekstrak Hedyotis diffusa Willd. dan doksorubisin mampu menimbulkan efek sinergis berdasarkan nilai CI.

English Abstract

Doxorubicin as chemotherapy for breast cancer is often limited due to its toxicity to the heart. Snake tongue grass (Hedyotis diffusa Willd.) is a herbal medicine prescribed in China as breast cancer treatment. Hedyotis diffusa Willd. and doxorubicin was combined to decrease doxorubicin’s dose used. In this study, we investigated IC50 value of Hedyotis diffusa Willd. and doxorubicin, their combination effects, and the concentrations that can have synergistic effects. Hedyotis diffusa Willd. was extracted using maceration method in 80% ethanol solvent. MCF-7 cell cultures were treated with 7 series concentrations for 24 hours, then IC50 values were measured using MTT assay. The concentrations used for combination assay were 1/2 IC50, 3/8 IC50, 1/4 IC50, and 1/8 IC50 then CI value was calculated. The IC50 value of Hedyotis diffusa Willd. was 930 ± 30.143 μg/ml (p<0.0001) and doxorubicin was 490 ± 83,293 nM (p<0.0001). The combination of Hedyotis diffusa Willd. and doxorubicin showed a synergistic effect on the concentration combination of doxorubicin was 61 nM and Hedyotis diffusa Willd. was 116 μg/ml and 232 μg/ml. Based on these results, the combination of Hedyotis diffusa Willd. and doxorubicin causes synergistic effects based on CI values.
