Indonesian Abstract

Kanker payudara merupakan penyebab umum kematian pada wanita di seluruh dunia. Rumput lidah ular (Hedyotis diffusa Willd.) dan kunyit (Curcuma domestica Val.) merupakan dua tanaman obat yang dipercaya dan telah diteliti sebagai terapi antikanker. Tujuan dari penelitian ini adalah membuktikan bahwa pemberian kombinasi ekstrak Hedyotis diffusa Willd. dan Curcuma domestica Val. mampu memberikan efek sitotoksik yang sinergis pada sel kanker payudara MCF- 7. Sel MCF-7 dipapar ekstrak dengan konsentrasi yang berbeda dan diinkubasi selama 24 jam. Uji sitotoksik dilakukan dengan metode MTT assay untuk mendapatkan nilai Inhibitory Concentration 50% (IC50). Kombinasi ekstrak Hedyotis diffusa Willd. dan Curcuma domestica Val. dengan empat dosis yang berbeda, yaitu 1/2 IC50, 3/8 IC50, 1/4 IC50, dan 1/8 IC50 sehingga dapat dilakukan perhitungan combination index (CI). Nilai IC50 yang diperoleh adalah 930 ± 52,199 μg/ml pada ekstrak Hedyotis diffusa Willd. dan 76 ± 15, 512 μg/ml pada ekstrak Curcuma domestica Val. Efek sinergis kuat didapatkan dari kombinasi ekstrak Hedyotis diffusa Willd. pada dosis 116 μg/ml dan Curcuma domestica Val. 10 μg/ml dengan nilai CI sebesar 0,297. Hasil penelitian menunjukkan bahwa kombinasi ekstrak Hedyotis diffusa Willd. dan Curcuma domestica Val. memiliki efek sinergis terhadap sel MCF-7 dan berpotensi sebagai agen kemopreventif pada kanker payudara.

English Abstract

Breast cancer is a common cause of death in women around the world. Snake tongue grass (Hedyotis diffusa Willd.) and turmeric (Curcuma domestica Val.) are two medicinal plants that have anticancer activity. The purpose of this study is to prove that combination of Hedyotis diffusa Willd. and Curcuma domestica Val. could perform a synergism cytotoxic effect against MCF-7 breast cancer cells. MCF-7 cells were treated with various concentrations of extract for 24 hours. The cytotoxic effect was performed by using MTT assay to obtain Inhibitory Concentration 50% (IC50) value. The combination of Hedyotis diffusa Willd. and Curcuma domestica Val. extract with four different doses, 1/2 IC50, 3/8 IC50, 1/4 IC50, and 1/8 IC50 can be calculated as combination index (CI). The value of IC50 obtained was 930 ± 52,199 μg/ml on Hedyotis diffusa Willd. extract and 76 ± 15, 512 μg/ml on Curcuma domestica Val. extract, respectively. The strong synergism effect was obtained from the combination of Hedyotis diffusa Willd. 116 μg/ml and Curcuma domestica Val. 10 μg/ml with the CI was 0,297. The results showed that the combination of Hedyotis diffusa Willd. and Curcuma domestica Val. extract has a synergism effect against MCF-7 cells and has potential an chemopreventive agent in breast cancer.
