Indonesian Abstract

Daun kemangi merupakan tanaman yang banyak dimanfaatkan dalam bidang pengobatan untuk mengobati berbagai penyakit seperti diare, perut kembung, disentri, dan kerusakan ginjal. Beberapa penelitian terdahulu mengungkapkan bahwa ekstrak daun kemangi mengandung senyawa fenol dan flavonoid yang mampu berperan sebagai antimikroba dan antioksidan. Metode yang umum digunakan untuk mendapatkan senyawa tersebut adalah maserasi. Namun, maserasi dinilai masih memiliki kelemahan seperti rendahnya jumlah senyawa metabolit sekunder yang dapat terekstrak. Guna meningkatkan hasil ekstraksi senyawa metabolit sekunder, metode maserasi dapat dikombinasikan dengan teknologi Pulsed Electric Field (PEF) sebagai perlakuan pendahuluan. Penelitian ini bertujuan untuk mengetahui pengaruh variasi kuat medan listrik dan durasi pretreatment terhadap jumlah senyawa fenol dan flavonoid dari ekstrak daun kemangi. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) dengan dua variabel, yaitu kuat medan listrik (2, 3, dan 4 kV/cm) dan durasi pretreatment (1, 2, dan 3 menit). Setiap kombinasi perlakuan diulang sebanyak tiga kali dan dihitung rata-ratanya. Hasil analisis sidik ragam menunjukkan bahwa variasi kuat medan listrik, durasi pretreatment, dan interaksi kedua variabel tersebut berpengaruh signifikan terhadap jumlah fenol dan flavonoid yang dihasilkan. Berdasarkan data perhitungan, jumlah fenol dan flavonoid tertinggi diperoleh pada kombinasi perlakuan kuat medan listrik 3 kV/cm selama 2 menit, yaitu 115,203±1,115 mg GAE/g ekstrak dan 75,816±0,723 mg QE/g ekstrak. Ekstrak dengan pretreatment PEF juga menunjukkan jumlah fenol dan flavonoid yang lebih tinggi dibandingkan perlakuan kontrol.

English Abstract

Basil leaf is one of the most well-known aromatic herb used for medicinal purposes to treat several diseases such as diarrhea, flatulence, nausea, dysentery, and kidney malfunction. Several previous scientific studies have revealed that basil leaf extract could act as an anti-microbial, antioxidant, anti-inflammatory, and cardiovascular agents. These actions are attributed to its composition, which is rich in phenol as well as flavonoids. So far, the common method to obtain both compounds is by conventional extraction method, namely maceration. However, maceration is considered to have weaknesses such as low yield of secondary metabolite compounds that can be extracted. In order to increase the extraction yield, the maceration method will be combined with Pulsed Electric Field (PEF) technology as pre-treatment. The aim of this study was to analyze the effect of electric field strength and pre-treatment time on the extraction of total phenol and flavonoid contents of basil leaves. This research used Randomized Block Design (RBD) with two variables, i.e electric field strength (2, 3, and 4 kV/cm) and pre-treatment time (1, 2, and 3 minutes). All the experiments were performed in triplicate and the results were averaged. Result of analysis of variance (ANOVA) showed that variation of electric field strength, pre-treatment time, and combination of both variables have significant effect on total phenol and flavonoid contents. The extract obtained using 3 kV/cm treatment for two minutes achieved the highest amounts of total phenols (115,203 ± 1.115 mg GAE/g extract) and flavonoid contents (75.816±0.723 mg QE/g extract). These treated samples also showed higher results compared to the untreated sample.
