Abstract

Pengendalian penyakit tanaman yang ramah lingkungan dapat dilakukan dengan aplikasi bakteri antagonis. Kelompok bakteri yang berpotensi sebagai agens antagonis dapat ditemukan di tanah, jaringan endofit dan air. Beberapa genus bakteri yang mampu berperan sebagai agens antagonis yaitu dari genus Bacillus, Pseudomonas, dan Pantoea. Di sumber air panas ditemukan bakteri Bacillus dan mempunyai manfaat dalam bidang pertanian karena dapat menghasilkan senyawa antibiotik. Sehingga penelitian ini bertujuan untuk mengetahui potensi bakteri dari sumber air panas sebagai agens antagonis terhadap patogen H. turcicum penyebab hawar daun pada tanaman jagung. Kegiatan penelitian ini dilaksanakan di Laboratorium Sentral Ilmu Hayati dan Laboratorium Penyakit Tumbuhan jurusan HPT, Fakultas Pertanian, Universitas Brawijaya, Malang dimulai pada bulan Juli hingga bulan Oktober 2018. Tahapan dari penelitian ini yaitu eksplorasi bakteri dari sumber air panas, seleksi bakteri, identifikasi bakteri, perbanyakan patogen H. turcicum, dan uji antagonis bakteri terseleksi terhadap patogen H. turcicum secara in vitro. Uji antagonis ini menggunakan Rancangan Acak Lengkap (RAL) dengan 8 perlakuan yaitu 7 perlakuan bakteri, dan 1 kontrol sebanyak 4 ulangan. Jika terdapat perbedaan nyata maka akan diuji lanjut menggunakan BNT pada taraf 5%. Variabel yang diamati dari penelitian ini yaitu karakterisasi bakteri antagonis dari sumber air panas dan daya hambat bakteri antagonis terhadap patogen H. turcicum. Hasil penelitian didapatkan 7 bakteri yang mampu menghambat pertumbuhan patogen H. turcicum. Pada pengujian antagonis secara in vitro, 7 perlakuan bakteri antagonis dan 1 kontrol menunjukkan hasil berbeda nyata. Zona hambat tertinggi yaitu pada bakteri S7 yang merupakan genus Bacillus sp. Hasil karakterisasi bakteri antagonis dengan kode isolat S1 termasuk kedalam genus Pantoea sp, bakteri S2 dan S7 termasuk kedalam genus Bacillus sp, bakteri S11, S14 dan S15 termasuk kedalam genus Erwinia sp, bakteri S16 belum teridentifikasi sehingga perlu dilakukan uji selanjutnya untuk mengetahui jenis genusnya.

English Abstract

Environmental-friendly control to plant disease can be done by antagonistic bacteria application. Groups of bacteria that are potentially antagonistic agents can be found in soil, endophytic tissue and water. Some genus of bacteria that are capable of acting as antagonistic agents were from the genus Bacillus, Pseudomonas, and Pantoea. Bacillus bacteria found in hot spring has benefits in agriculture because it can produce antibiotic compounds. So the aim in this study is to determine the potential of bacteria from hot spring as antagonistic agents against H. turcicum pathogen that cause leaf blight in corn plants. The research activity was carried out at the Central Laboratory of Life Sciences and the Laboratory of Plant Diseases majoring in HPT, Faculty of Agriculture, Brawijaya University, Malang starting from July until October 2018. The stages of this research are bacterial exploration from hot spring, bacteria selection, bacteria identification, multiplication of pathogen H. turcicum, and antagonist tests on selected bacteria against H. turcicum in vitro. This antagonist test uses a completely randomized design (CRD) with 8 treatments that is 7 bacterial treatments with 1 control and 4 times repeated. If there are significant differences, it will be tested further using LSD at the level of 5%. The variables observed from this study were the characterization of antagonistic bacteria from hot spring and bacteria inhibition against H. turcicum pathogen. The results of this study is 7 bacteria were found capable to inhibit the growth of H. turcicum pathogen. In antagonist test in vitro, 7 treatments of antagonistic bacteria and 1 control showed different results. The highest inhibition zone is on the S7 bacteria that is identified as Bacillus sp. The results of characterization antagonistic bacteria with code S1 isolate included in to genus Pantoea sp, S2 and S7 bacteria included in to genus Bacillus sp, S11, S14 and S15 bacteria included in to genus Erwinia sp. S16 bacteria has not been identified so that further testing is needed to show the genus of the bacteria.
