Indonesian Abstract

Pola makan tak seimbang seperti diet tinggi lemak dapat menimbulkan efek buruk bagi kesehatan, antara lain peningkatan radikal bebas dalam tubuh yang disebut juga stres oksidatif hingga terjadi inflamasi kronis yang disebabkan paparan radikal bebas terus-menerus dan mengakibatkan kerusakan jaringan dan ketidakseimbangan antioksidan endogenous (enzimSuperoksida DismustaseatauSOD) dalam melawan paparan radikal bebas tersebut.Salah satu komoditas pangan dari perairan yang dikenal memiliki aktivitas antioksidan dan antiinflamasi, serta belum banyak dimanfaatkan yaitu algaEucheuma cottonii (alga merah) dan Sargassum sp.(alga cokelat).Tujuan dari penelitian ini yaitu mengetahui pengaruh konsumsi diet tinggi lemak dalam menginduksi stres oksidatif yang ditandai peningkatan malondialdehid (MDA) dan penurunan SOD serta mengetahui pengaruh serbuk algaE. cottoniidan Sargassum sp. terhadap kadar MDA dan SOD serum darah tikus wistar yang diinduksi 50% diet tinggi lemakyang berasal dari 10% minyak kedelai dan 40% lemak sapi. Pada penelitian ini akan diamati efek pemberian serbuk E. cottonii dan Sargassum sp.dalam pakan tikus wistar terhadap kadar MDA dan SOD serum darah akibat konsumsi diet tinggi lemak selama 4 minggu. Penelitian dilakukan menggunakanRancangan Acak Lengkap (RAL) dimana respon diperoleh dari 4 perlakuan pakan, yaitu pakan standar, pakan tinggi lemak, pakan tinggi lemak dengan serbuk E. cottoniidan pakan tinggi lemak dengan serbuk Sargassum sp. Pada akhir percobaan, tikus dibius dan darah dikumpulkan dari jantung untuk analisis MDA serta SOD serum. Hasil penelitian menunjukkan, kadar total fenol serbuk E. cottonii 158,08 ± 28,47mg PGE/ g sedangkan pada serbuk Sargassum sp. 180,91 ± 13,51mg PGE/ g. Pemberian serbuk E.cottonii dan Sargassum sp. tidak berpengaruh nyata pada kadar MDA serum tikus yang diinduksi stres oksidatif dengan diet tinggi lemak masing-masing sebesar2,51± 0,11 dan 2,57± 0,27ng/100 μL. Pemberian serbuk E.cottonii dan Sargassum sp. juga tidak berpengaruh nyata pada kadar SOD serum tikus yang diinduksi stres oksidatif dengan diet tinggi lemak masing-masing sebesar 24,32± 5,41dan 18,14 ± 5,11unit/100 μL. Hal ini menandakan serbuk E. cottonii dan Sargassum sp.pada dosis percobaan masing-masing 194,77 g/kg pakan dan 151,20 g/kg pakan belum mampu menurunkan kadar MDA dan meningkatkan kadar SOD pada tikus yang diinduksi stres oksidatif melalui pemberian pakan tinggi lemak.

English Abstract

Unbalanced diets such as a high-fat diet has been reported to cause adverse effects on health, such as an elevatedfree radical that lead to chronic inflammation and trigger tissue damage. One of the emerging food marine biosources used as antioxidants and anti-inflammatory agent is algae. Eucheuma cottonii (red algae) and Sargassum sp. (brown algae) were reported to have high antioxidant activity due to polyphenolic compounds. The aim of this study was to investigate the effect of a high-fat diet in inducing oxidative stress characterized by malondialdehyde (MDA) and decreased Superoxyde Dismustase (SOD) and to investigate the effect of algae powder E. cottonii and Sargassum sp. on MDA and SOD levels in oxidative stressinduced rats with 50% high-fat diets composed of 10% soybean oil and 40% tallow. Rats were randomly assigned into 4 groups differing in the type of diet, standard with normal diet, high fat diet, high fat diet added with E. cottonii powder and high fat diet added with Sargassum sp. powder for 4 weeks (28 days). At the end of the experiment, rats were euthanized and blood was collected by means of cardiac puncture for serum MDA and SOD analyse. The results showed that total phenolic compounds in E. cottonii and Sargassum sp. powderwere158,08 ± 28,47mg PGE/ g powder and180,91 ± 13,51 mg PGE/ g powder respectively. Animal study showed that both algae powderdid not affect MDA level in serum of oxidative stress induced rats with high fat dietresulting2,51± 0,11and 2,57 ± 0,27ng/100 μL of MDA levels respectively. The MDA levels of rats treated with both algae did not differ from those not treated with algae (2,67± 0,39 ng/100 μL), and higher than those given a standard normal diet (1,38± 0,18 ng/100 μL).Furthermore, both algae powder were not able to increase SOD level in serum of oxidative stress induced rats resulting24,32± 5,41and 18,14 ± 5,11unit/100 μL SOD level respectively. The SOD levels did not differ with those not treated with algae (16,79± 6,48 unit/100μL) and lower than those given a standard normal diet (46,43± 4,67 unit/100μL). In the present study, it can be concluded that the dose of E. cottonii and Sargassum sp. powder 194,77 g/kg feed dan 151,20 g/kg feed usedrespectively were not able to decrease MDA serum level neither did increase SOD serum level in oxidative stress induced rats with high fat diet.
