Indonesian Abstract

Fibrosis hepar merupakan salah satu akibat dari kerusakan hepar kronik yang ditandai dengan akumulasi berlebihan protein Extracellular Matrix (ECM) termasuk kolagen dengan gejala yang muncul seiring bertambahnya tingkat keparahan fibrosis. Pembuatan hewan model fibrosis hepar dilakukan dengan menggunakan hewan coba yang diinduksi CCl4. Penelitian ini dilakukan untuk mengetahui indikasi ekstrak gambir (Uncaria gambir Roxb.) sebagai alternatif tindakan preventif pada hewan model tikus fibrosis hepar berdasarkan kadar albumin dan bilirubin total. Penelitian ini menggunakan hewan coba tikus jantan berumur 8-12 minggu dengan berat 150-200 gram, Rancangan Acak Lengkap (RAL) menggunakan lima kelompok, kontrol positif (CCl4), kelompok kontrol negatif, kelompok preventif dosis 130 mg/kg BB (I), 260 mg/kg BB (II), dan 390 mg/kg BB (III). Pengukuran kadar albumin dan kadar bilirubin total menggunakan spektofotometer dan dianalisis menggunakan ragam one-way ANOVA dan dilanjutkan dengan uji Tukey alfa=5%. Hasil dari penelitian ini menunjukkan bahwa pemberian ekstrak gambir sebagai preventif pada tikus model fibrosis hepar dengan dosis efektif 390 mg/kg BB dapat meningkatkan kadar albumin serum dan menurunkan kadar bilirubin total dari kontrol positif. Kesimpulan penelitian ini bahwa ekstrak gambir dapat digunakan sebagai preventif fibrosis hepar pada tikus yang diinduksi karbon tetraklorida

English Abstract

Hepatic fibrosis is one result of chronic liver damage characterized by excessive accumulation of Extracellular Matrix (ECM) proteins including collagen with symptoms that appear with increasing severity of fibrosis. Animal model making of hepatic fibrosis was performed using CCl4 induced animal. This study was conducted to determine the indications of gambir extract (Uncaria gambir Roxb.) As an alternative preventive action in mouse model of liver fibrosis based on total albumin and bilirubin levels. The study used male rats from 8 to 12 weeks old with weight 150-200 grams, Completely Randomized Design (CRD) using five groups, positive control (CCl4), negative control group, dose preventive group 130 mg/kg body weight (I), 260 mg/kg BW (II), and 390 mg/kg BW (III). Measurements of albumin content and total bilirubin content were measured using a spectrophotometer and analyzed by variety one-way ANOVA and followed by testing Tukey alfa = 5%. The results of this study indicate that giving gambir extract as preventive in mouse model of hepatic fibrosis with effective dose of 390 mg/kg BW can increase serum albumin level and decrease total bilirubin level from positive controls. The conclusion of this research that gambir extract can be used as a preventive liver fibrosis in rats induced by carbon tetrachloride.
