Indonesian Abstract

Telah dilakukan analisis pengaruh penyinaran sinar gamma (γ) terhadap kadar insulin pankreas sebelum dan setelah pemberian ekstrak buah pare ( Momordica charantia L.) pada hewan coba mencit ( Mus musculus ) yang dibebani glukosa. Tujuan dari penelitian ini adalah menganalisi s efek radiasi sinar gamma terhadap ketahanan organ pankreas dalam memproduksi insulin dan mengkaji efek pemberian ekstrak buah pare ( Momordica charantia L . ) terhadap ketahanan organ pankreas dalam memproduksi insulin pada mencit yang diradiasi sinar gamma. Sinar gamma merupakan radiasi pengion yang dapat memicu terjadinya kerusakan sel bahkan kematian sel apabila berinteraksi dengan sel. Kerusakan sel akibat radiasi berkaitan erat dengan radikal bebas yang dihasilkan ketika radiasi berinteraksi dengan sel. Mekanisme pertahanan sel terhadap radikal bebas melibatkan suatu senyawa aktif yang disebut antioksidan. Antioksidan diketahui berfungsi menetralkan dan menangkap radikal bebas serta mencegah proses oksidasi. Sebanyak 180 ekor mencit jantan digunakan dalam penelitian ini dan dibagi menjadi 4 kelompok ; kontrol (K-), radiasi ( R- ), ekstrak buah pare tanpa radiasi ( K+ ) dan ekstrak buah pare dengan radiasi (R+). Mencit diberi kan ekstrak buah pare peroral dengan dosis 200 mg/kg BB (EP1), 400 mg/kg BB (EP2), 600 mg/kg BB (EP3), 800 mg/kg BB (EP4) dan 1000 mg/kg BB (EP5) selama dua minggu sebelum dan selama proses radiasi dengan sinar gamma. Proses radiasi dilakukan secara fraksinasi selama lima hari dengan 100 rad per fraksi mulai dari 100 rad, 200 rad, 300 rad, 400 rad sampai 500 rad tiap perlakuan . Darah diambil melalui jantung mencit, selanjutnya disentrifugasi se h ingga diperoleh serum darah yang akan diuji ELISA lebih lanjut untuk mengetahui kadar insulinnya. Hasil penelitian menunjukkan bahwa pemberian radiasi mampu menurunkan kadar insulin bila dibandingkan dengan control , dimana kadar insulin cenderung mengalami penurunan seiring dengan penambahan dosis radiasi. Pemberian ekstrak buah pare mampu meningkatkan kadar insulin bila dibandingkan dengan kontrol, dimana kadar insulin cenderung meningkat seiring dengan penambahan dosis ekstrak. P erlakuan ekstrak buah pare juga mampu menekan penurunan bahkan mempertahankan kadar insulin akibat paparan radiasi. B erdasarkan perbandingan rata-rata kadar insulin antar kelompok perlakuan ekstrak buah pare, dosis EP5 ( 1000 mg/kg BB ) merupakan dosis yang paling efektif untuk meningkatkan kadar insulin serum mencit, menekan penurunan dan mempertahankan kadar insulin akibat paparan radiasi.

English Abstract

It has been analysed effects of gamma radiation exposure of pancreas insulin levels in glucose-loaded mice ( Mus muscullus ) before and after treated with Bitter guard`s fruit ( Momordica charantia L.) extracts . The objectives of this research were to analyze effects of gamma radiation exposure in mice (Mus muscullus) pancreas resistance to produce insulin after exposured. The research was also aimed to investigate effects of treated with bitter guard`s fruit ( Momordica charantia L.) extract in mice ( M us muscullus ) pancreas resistance to produce isulin after exposured. Gamma rays are ionizing radiation causes cells damage even death when them interacted with the cells. Cells damage caused by radiation exposure related to free radicals as a product when the interaction happened. The cell defense mechanisms againts free radicals involve an active compound called antioxidant. This compounds can react with free radicals and neutralize it, and prevent the oxidation. One hundred eighty mice were used in this research. They were divided into four group : control (K-), radiation ( R- ), bitter guard`s fruit extract without radiation ( K+ ) and bitter guard`s fruit extract with radiation (R+). The Mice were treated with 200 mg/kg of body weight (EP1), 400 mg/kg of body weight (EP2), 600 mg/kg of body weight (EP3), 800 mg/kg of body weight (EP4), and 1000 mg/kg of body weight (EP5) orally of Bitter Guard`s fruit extract for two weeks before and during the irradiation. Exposure of g amma ray radiation is given fractionally for five days with 100 rad per fraction ranging from 100 rad, 200 rad, 300 rad, 400 rad and 500 rad per treatment. Serum insulin levels was measured by ELISA method by us ing the blood serum of mice, where the blood samples were taken from it heart than centrifuged to obtain the blood serum. The results showed that gamma radiation exposure decreasing insulin levels, tended to decrease from control. The decrese of insulin level was proportional to gamma radiation dosage. Treated with bitter guard`s fruit extract increasing insulin level , tended to increase from control. The increase of insulin levelwas proportional to bitter guard`s fruit extract dosage. The bitter guard`s fruit extract also supressing the decease and maintaining insulin level s caused by gamma radiation exposure. Comparison of average insulin level among dosage groups of bitter guard`s extract and group of control, showed that EP5 dosage (1000 mg/kg body weight) is the most effective to increase serum insulin level s of mice, supressing the decrease and maintaining of insulin level s caused by gamma radiation exposure.
