English Abstract

Typhoid fever is a serious and often fatal disease in developing countries. It is caused by Salmonella Typhi . Obviously, Salmonella Typhimurium infection in mice is widely accepted as an experimental model for typhoid fever in humans. interleukin (IL-12) family of cytokines is potential to treat immune-mediated inflammatory diseases. IL-12 induces interferon-γ (IFN-γ). Date fruit (Phoenix dactilifera) and its constituents show a role in diseases prevention through anti-oxidant, anti-inflammatory, and anti-bacterial activity. This study used 25 mice divided into 5 groups, including control negative (without infection), control positive (infected with S. Typhimurium), treatment group: P1 ( 100mg/kgBW), P2 (200mg/ kgBW), and P3 (400mg/ kgBW). ELISA was used to measure IL-12 and IFN-γ levels and culture was used to measure bacterial colonies in liver. results indicate that IFN-γ levels decreased significantly at dose 400 mg/kgBW compared with C+ (p 0.05), but descriptively, level of IFN-γ increased at dose 100mg/kgBW. In addition, IL-12 levels increased significantly at dose 400 mg/kgBW compared with (p 0.05), but it decreased at dose 100mg/kgBW. bacteria colonies in liver sample of mice induced S.Typhimurium bacteria in positive control group become zero after treatment was givenin form of phoenix dactylifera (p1.100mg/kg, p2. 200mg/kg ,p3. 400mg/kg ) (p0.05). re was no bacteria found in all treatment groups. In conclusion, ethanol extract of Phoenix dactilifera can improve immune response by increasing IL-12 level and decreasing IFN-γ level, as well as inhibiting systemic diseases by killing bacteria colony in liver.
