Abstract

Pengendalian hayati populasi nyamuk Aedes aegypti sering dilakukan menggunakan Bacillus thuringiensis. Banyak produk Bacillus thuringiensis dalam bentuk formula padat dan cair yang dapat ditemukan di dunia, antara lain MOSNONTM yang diproduksi oleh salah satu industri di Jepang. Tujuan penelitian ini adalah menganalisis toksisitas isolat MDN1TK2 (Bacillus thuringiensis asal Madiun) dan Bacillus thuringiensis D-142 (MOSNONTM) dalam membunuh larva Aedes aegypti instar III. Metode yang digunakan meliputi rearing Aedes aegypti, isolasi isolat Bacillus thuringiensis D-142, peremajaan isolat MDN1TK2 dan Bacillus thuringiensis D-142, pewarnaan Gram dan endospora, uji toksisitas isolat MDN1TK2 dan Bacillus thuringiensis D-142, serta analisis data menggunakan SPSS for Windows release 16.0. Uji toksisitas dilakukan pada gelas plastik yang diisi air sumur dan kultur bakteri dengan konsentrasi 90 %, 80 %, 70 %, 60 %, 50 %, 15 %, 10 %, 5 %, 0.5 %, 0.01 %, 0.0002 %, 0.000004 %, dan 0 %. Volume total tiap gelas plastik sebesar 50 mL dan ditambahkan 20 larva Aedes aegypti instar III. Jumlah kematian larva Aedes aegypti diamati pada 24, 48, 72, dan 96 jam. Parameter yang diamati selama uji toksisitas adalah mortalitas larva Aedes aegypti, suhu, dan pH air. Nilai LC50 ditentukan dengan analisis Probit. Hasil penelitian menunjukan bahwa nilai LC50-96jam untuk isolat MDN1TK2 sebesar 5,9×109 sel/mL sedangkan untuk isolat Bacillus thuringiensis D-142 sebesar 3×102 sel/mL.

English Abstract

Biological control for supressing Aedes aegypti populations was investigated using Bacillus thuringiensis. Many kind of Bacillus thuringiensis product already found in the world, for example MOSNONTM which is produced by Japan industry. The goal of this study was to determine the toxicity of the MDN1TK2 isolates (Bacillus thuringiensis was isolated from Madiun) and Bacillus thuringiensis D-142 (MOSNONTM) for killing the third instar larvae of Aedes aegypti. Methods that already conducted consist of rearing of Aedes aegypti, renewal for MDN1TK2 isolates and Bacillus thuringiensis D-142 isolates, Gram and endospores staining, toxicity test of Bacillus thuringiensis D-142 and MDN1TK2 isolates, and also analyzed data using SPSS for Windows release 16.0. Toxicity test was carried on plastic cups filled with wells water and bacterial cultures for different concentrations. The concentration of Bacillus thuringiensis 90 %, 80 %, 70 %, 60 %, 50 %, 15 %, 10 %, 5 %, 0.5 %, 0.01 %, 0.0002 %, 0.000004 %, and 0 %. Each plastic cup was added by 20 Aedes aegypti larvae (third stage). The number of Aedes aegypti larvae were observed for different times (24, 48, 72, and 96 hours). Mortality number of Aedes aegypti, water temperature, pH of water were observed during the research. Determination of LC50 value was done by Probit analysis. The result showed that LC50-96h of MDN1TK2 isolates was 5,9×109 cell/mL and Bacillus thuringiensis D-142 isolates was 3×102 cells/mL.
