Abstract

Kitosan merupakan polisakarida rantai lurus yang tersusun atas monomer glukosamina dengan ikatan glikosidik. Tujuan penelitian ini adalah melakukan isolasi kitosan dari cangkang rajungan dan mengoptimalisasi reaksi deasetilasinya, mempelajari reaksi substitusi kitosan dan kurkuminoid, serta mengetahui kestabilan produk hybride kitosan-kurkuminoid dalam suasana asam dan basa. Reaksi deasetilasi kitosan dilakukan dengan dua variasi, yaitu variasi konsentrasi basa (NaOH 50%, NaOH 60%, NaOH 70%) dan variasi lama reaksi deasetilasi (9, 16, 24 jam). Hasil isolasi kitosan dari cangkang rajungan berbentuk serbuk berwarna coklat muda dengan rendemen 46,25%. Nilai DD (Derajat Deasetilasi) tertinggi dari hasil deasetilasi kitosan pada konsentrasi NaOH 70% dengan lama reaksi deasetilasi 24 jam, yaitu sebesar 87,96%. Sedangkan reaksi substitusi kitosan-kurkuminoid juga dilakukan dengan dua variasi, yaitu variasi rasio kitosan-kurkuminoid (1:1, 1:2, 1:4) (b/b) dan variasi waktu reaksi (0, 30, 60, 90, 120 menit) dalam suasana asam dan basa. Hasil reaksi dengan variasi rasio kitosan-kurkuminoid memberikan data nilai DS (Derajat Substitusi) tertinggi, yaitu sebesar 0,578 pada rasio kitosan-kurkuminoid 1:4 (b/b). Lamanya waktu reaksi dengan metode konvensional menggunakan pengaduk magnetik baik dalam suasana asam maupun basa berpengaruh pada nilai absorbansi kurkuminoid sisa hasil reaksi.

English Abstract

Chitosan is a straight chain polysaccharide composed of glucosamine monomers with glycoside linkage. This research was purposed to isolate chitosan from crab shell, optimization of deacetylation reaction, and to study the substitutions reaction of chitosan and curcuminoid. And also to know the stability of the product hybride chitosan-curcuminoid in acid and base condition. Deacetylation reaction was performed with two variations, that is variation of base concentration (NaOH 50%, NaOH 60%, NaOH 70%) and variation of deacetylation reaction times (9, 16, 24 hours). It was found that chitosan was isolated from crab shell as a brown powder in 46,25% yield. Moreover, deacetylation reaction of chitosan provided DD (Degree of Deacetylation) value 87,96% when using 70% of NaOH solution for deacetylation reaction time of 24 hours. While the substitution reaction of chitosan-curcuminoid also performed with two variations, that is the variation of the ratio of chitosancurcuminoid (1:1, 1:2, 1:4) (b/b) and reaction time (0, 30, 60, 90, 120 minutes) in acid and base condition. Reaction products with variation in the ratio of chitosan-curcuminoid provide the highest DS (Degree of Substitution) value, that is equal to 0,578 for ratio chitosancurcuminoid 1:4 (b/b). The length of reaction time due to the conventional method using magnetic stirring both in acid or base condition take effect on the absorbance value of curcuminoid residual reaction product
