Abstract

Kakao merupakan salah satu hasil perkebunan terbesar di Indonesia. Pada umumnya kakao diambil bijinya untuk diolah dan diekspor, sehingga 75% sisanya berupa pod kakao menjadi limbah yang belum optimal pengolahannya untuk dimanfaatkan menjadi produk lain bernilai lebih. Pod kakao mengandung lignoselulosa, terdiri dari selulosa 30,24%; hemiselulosa 6,66%; dan lignin 38,45%. Berdasar komposisi kimia, limbah pod kakao yang melimpah dapat dimanfaatkan menjadi produk lain. Pengolahan pod kakao memerlukan pretreatment untuk mempermudah mendegradasi struktur lignoselulosa, maka digunakan pretreatment biologi dalam penelitian ini menggunakan jamur pelapuk coklat Serpula lacrymans. Salah satu faktor yang mempengaruhi pertumbuhan jamur adalah nutrisi untuk mempercepat pertumbuhan dan proses degradasi. Nutrisi yang ditambahkan adalah glukosa dan CaSO4, karena nutrisi tersebut berpengaruh mempercepat pertumbuhan jamur dalam memproduksi fenol. Penelitian ini menggunakan Response Surface Methode (RSM) dan didapatkan hasil solusi optimal dari penambahan glukosa 1,00% (g/l) dan CaSO4 0,25% (g/l) yaitu TSP = 0,163 mg/g, TGR = 183,375 mg/g, nilai pH = 5,86, dan susut berat = 0,011 atau 1,1%, dengan desirability = 0,609. Nilai TSP yang mendekati solusi optimal yaitu 0,169 mg/g (F1W3) dipilih untuk diuji menggunakan LC MS/MS. Hasil uji LC MS/MS menunjukkan adanya dua komponen senyawa fenolik.

English Abstract

Cacao is one of the largest plantation crops in Indonesia. In general, taken cacao seeds to be processed and exported, to 75% in the rest of the cacao pod becomes waste that is not optimal processing to be used to produce other products worth more. Cacao pod contains lignocellulose, composed of cellulose 30.24%; hemicellulose 6.66%; and lignin 38.45%. Based on the chemical composition, abundant cacao pod waste can be utilized as another product. Processing cacao pod require pretreatment to facilitate degrade lignocellulose structure, then used a biological pretreatment in this research using a brown rot fungi Serpula lacrymans. One of the factors that influence the growth of the fungus is nutrient to accelerate growth and degradation processes. The added nutrients are glucose and CaSO4, because these nutrients affect accelerate the growth of fungi in producing phenol. This study using Response Surface Method (RSM) and the optimal solution is obtained from the addition of glucose to 1.00% (g/l) and CaSO4 0.25% (g/l), they are TSP = 0.163 mg/g, TGR = 183.375 mg/g, pH = 5.86, and weight loss = 0.011, or 1.1%, with the desirability = 0.609. TSP sample Values approaching the optimal solution is 0.169 mg/g (F1W3) have to be tested using a LC MS/MS. The test results LC MS/MS showed two components of phenolic compounds.
