Indonesian Abstract

Diazinon merupakan salah satu insektisida golongan organophosphate yang bekerja di dalam tubuh hewan dengan jalan menghambat molekul acetylcholinesterase (AChE) secara kompetitif dan menyebabkan peningkatan produksi Reactive Oxygen Species (ROS). Molekul ROS akan mempengaruhi aktivitas enzim-enzim antioksidan dalam tubuh seperti enzim Superoxide Dismutase (SOD) dan dapat mengoksidasi komponen sel seperti lipid, mengakibatkan kerusakan pada sel dan jaringan. Tujuan penelitian ini yaitu untuk mengetahui pengaruh toksisitas organophosphate (diazinon) terhadap gambaran histopatologi duodenum dan aktivitas Superoxide Dismutase (SOD) dalam serum tikus putih (Rattus norvegicus). Tikus dibagi dalam 4 kelompok perlakuan yaitu kelompok kontrol negatif (K-), kelompok P1, P2, dan P3 diberikan diazinon secara per-oral selama 8 minggu dengan dosis berturut-turut 20 mg/kgBB, 40 mg/kgBB, dan 60 mg/kgBB. Pengukuran aktivitas SOD serum menggunakan metode spektofotometri, dan perubahan histopatologi duodenum menggunakan pewarnaan HE. Data aktivitas SOD dianalisa secara statistik dengan one-way ANOVA dan uji lanjutan dengan uji Tukey α=0,05, sedangkan gambaran histopatologi duodenum dianalisis secara kualitatif. Hasil penelitian menunjukkan bahwa pemberian diazinon dengan dosis 20 mg/kgBB, 40 mg/kgBB, dan 60 mg/kgBB secara signifikan (p<0,05) mampu menurunkan aktivitas SOD dan menyebabkan kerusakan pada vili duodenum berupa hiperplasia sel epitel, erosi epitel, degenerasi melemak, hiperplasia sel kelenjar Liberkuhn dan hemoragi pada lamina propria. Kesimpulan dari penelitian ini yaitu dengan dosis pemberian diazinon terendah (20 mg/kgBB) telah menimbulkan efek toksik pada tikus putih (Rattus norvegicus) yang ditandai dengan penurunan aktivitas SOD sebesar 20,59% dan kerusakan vili duodenum.

English Abstract

Diazinon is an organophosphate insecticide, that acts upon an animal’s body. The Diazinon can inhibit acetylcholinesterase (AChE) competitively, and increase the production of Reactive Oxygen Species (ROS). ROS affect the activity of antioxidant enzymes in the body such as superoxide dismutase (SOD), and at the same time ROS oxidize cell components such as lipids, causing damage to cells and tissues. The purpose of this study was to determine the toxicity effect of organophosphate (Diazinon) towards duodenum histopathology and the activity of SOD serum in rats (Rattus norvegicus). Rats were divided into 4 groups, namely the negative control groups (K-), the group P1, P2, and P3 were given diazinon for 8 weeks orally with dose of 20 mg/kgBW, 40 mg/kgBW, and 60 mg/kgBW respectively. The activity of SOD serum were measured by using the spectrophotometry and histopathological changes of duodenum was observed using HE staining. The activity of SOD serum were analyzed statistically using one-way ANOVA and followed by Tukey test with α=0.05, while histopathological changes in duodenum were analyzed qualitatively. The results of this research indicated that administration of diazinon orally with dose of 20 mg/kgBW, 40 mg/kgBW, and 60 mg/kgBW significantly decreased SOD activity and caused damage of duodenal villi such as hyperplasia of epithelial cells, epithelial erosion, fatty degeneration, hyperplasia of Liberkuhn gland cells, and hemorrhage in the lamina propria. The conclusion is the lowest doses of diazinon (20 mg/kgBW) caused toxic effect in white rats (Rattus norvegicus) which shown by the decrease of SOD activity 20.59% and damage of duodenal villi.
