Indonesian Abstract

Hiperkolesterolemia sering terjadi pada hewan karena pola pemberian pakan berlemak dan kadar kolesterol tinggi yang melebihi kebutuhan tubuh. Metabolisme hiperkolesterol menjadi asam empedu menghasilkan radikal bebas mengakibatkan kerusakan jaringan hepar dan meningkatkan ekspresi TNF-α. Pencegahan kerusakan jaringan hepar akibat hiperkolesterolemia dapat dengan memberikan arang aktif. Interaksi antara kolesterol dengan karbon aktif terjadi adsorpsi secara fisika. Penelitian ini bertujuan untuk mengetahui pengaruh terapi preventif arang aktif terhadap ekspresi TNF-α hati dan histopatologi hati tikus (Rattus norvegicus) yang diinduksi hiperkolesterol. Penelitian ini menggunakan tikus putih (Rattus norvegicus) strain Wistar dibagi dalam 6 kelompok perlakuan yaitu: Kelompok negatif, kelompok positif diberi diet hiperkolesterol sebanyak 3,02 g/ekor, kelompok perlakuan diberi diet hiperkolesterol dan terapi arang aktif 340 mg/ekor, 680 mg/ekor, dan 1020 mg/ekor serta perlakuan hanya diberi arang aktif 680 mg/ekor diberikan peroral mulai hari ke 8-21 hari. Ekspresi TNF-α organ hati diamati dengan metode IHK dibantu software Immunoratio, dianalisa dengan uji ANOVA dilanjutkan uji Tukey (p<0,01). Gambaran histopatologi hepar diamati secara kualitatif dengan pewarnaan HE. Hasil penelitian pemberian arang aktif dapat menghambat perlemakan hepar dan menghambat kenaikan TNF-α sebesar 79,19% dengan dosis pemberian efektif 680 mg/ekor setelah pemberian diet hiperkolesterol. Kesimpulan penelitian ini arang aktif dapat mencegah hiperkolesterolemia berdasarkan penghambatan kenaikan TNF-α dan penghambatan histopatologi hepar.

English Abstract

Hiperkolesterolemia often occurs in animals because of the feeding patterns of fatty and high cholesterol levels that exceed the needs of the body. The metabolism cholesterol to be bile acid produces free radicals induce tissue damage and improves expression TNF-α of liver. Active charcoal can be prevention of liver tissue damage. Interaction between cholesterol with activated charcoal occurs adsorption in physics. This research aims to know the effect of preventive therapy against active charcoal expression of TNF-α and histopathology liver rat (Rattus norvegicus) induced hiperkolesterol. This research uses the white rat (Rattus norvegicus) strain Wistar which were divided into 6 groups of treatment: negative group, positive group were given diet of hipercholesterol as much as 3.02 g/rat, the group treatment was given diet hipercholesterol 3.02 g/rat and therapy active charcoal 340 mg/rat, 680 mg/rat, and 1020 mg/rat and treartmen with 680 mg/rat active charcoal only, given peroral 8-21 day. Expression of TNF-α liver was observed with immunohistochemistry helped by software imunnorati, analyzed with ANOVA test and followed by Tukey test (p < 0.01). The description of the histopathology liver observed qualitatively with HE coloring. Result of this research, the granting of active charcoal can inhibit fatty of liver and inhibit 79,19% the expression of TNF-α by granting effective dose 680 mg/rat. The conclusion is active charcoal can prevent the hipercholesterolemia based on the inhibition of TNF-α and increases the inhibition of histopathology liver.
