Indonesian Abstract

Asma adalah gangguan sistem respirasi yang disebabkan oleh perubahan kepekaan sistem imun saluran respirasi yang disebabkan oleh alergen. Asma mengakibatkan perubahan struktur dan kimia pada percabangan tracheobronkial. Asma dapat diperparah oleh infeksi rongga mulut akibat paparan Lipopolisakarida (LPS) dari bakteri Gram negatif. Omega-3 memiliki aktivitas antiinflamasi. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh omega-3 pada tikus asma ditinjau dari kadar MDA dan ekspresi TNFα bronkus tikus asma. Tikus model asma disiapkan dengan sensitasi alergi menggunakan Ovalbumin secara intraperitonial dan inhalasi serta pemberian LPS Phorphyromonas gingivalis secara intrasulkuler. Penelitian ini menggunakan lima kelompok tikus, yaitu kelompok kontrol negatif, kontrol positif (asma), serta terapi omega-3 dosis 30 mg/ekor/hari, 50 mg/ekor/hari, dan 70 mg/ekor/hari. Kadar MDA diukur dengan metode Thiobarbaturic Acid (TBA) sedangkan ekspresi TNFα diamati menggunakan metode Immunohistokimia. Data dari hasil perlakukan dianalisa menggunakan Microsoft Office Exel dan SPSS untuk Windows menggunakan analisis ragam ANOVA dan uji lanjutan Beda Nyata Jujur (BNJ) α = 5%. Hasil penelitian menunjukkan pemberian terapi omega-3 secara signifikan (p<0,05) mampu menurunkan kadar MDA dengan penurunan tertinggi sebesar 68,39% dan menurunkan ekspresi TNFα bronkus tikus model asma dengan penurunan tertinggi sebesar 56,4% pada dosis terapi 70 mg/ekor/hari. Kesimpulan dari penelitian ini adalah omega-3 berpotensi sebagai bahan terapi asma.

English Abstract

Asthma is a respiratory disorder caused by altered immunosensitivity of the respiratory tract due to inhaled allergens. This hyperresponsiveness causes structural and chemical changes of tracheobronchial. The symptoms of asthma become more severe by oral infections of lipopolysaccharide (LPS) of Gram-negative bacteria. Omega-3 has anti-inflammatory activity. The purpose of this study was to determine the therapeutic effect of omega-3 as anti-inflammatory on asthma rats based on the Malondialdehyde (MDA) level and Tumor Necrosis Factor Alpha (TNFα) expression on bronchial asthma rats. Asthma rats were prepared by allergent sensitization of ovalbumin administered by intraperitonial injection and inhalation, also intrasulcular injection of LPS from Phorphyromonas gingivalis. Five groups of rats (Rattus norvegicus) were used in this study: control group, asthma group, and three groups of omega-3 therapy dose of 30; 50; and 70 mg rat/day. Malondialdehyde levels were determined using Thiobarbaturic Acid (TBA) assay and expression of TNFα were observed using immunohistochemistry. The data was analyzed with Microsoft Office Exel and SPSS for Windows with ANOVA and continued with Tukey test. The result showed that omega-3 therapy decreased MDA level of bronchial’s asthma rats significantly (P<0.05) with the best derivation of 68.39%, and also decreased TNFα expression with the best result of 65.4%. The conclusion of the research was omega 3 had potentially to be used as theraphy for astma.
