Indonesian Abstract

Buncis (Phaseolus vulgaris L.) merupakan salah satu komoditas yang cukup popular di masyarakat. Buncis dapat berkhasiat untuk mencegah dan mengobati Diabetes Mellitus (Amin, 2014). Produksi buncis di Indonesia pada tahun 2011, 2012, dan 2013 mengalami fluktuasi, yaitu 334.659 ton, 322.145 ton menjadi 327.378 ton (Anonymous, 2014). Dalam upaya perbaikan kualitas dan produktivitas dari tanaman buncis, telah dilakukan perakitan varietas baru yaitu dengan melakukan persilangan antara varietas buncis introduksi (Purple Queen) dengan varietas lokal (Gogo Kuning, Gilik Ijo, Mantili). Persilangan tersebut bertujuan untuk memperoleh keturunan berdaya hasil tinggi dan berpolong ungu. Pada generasi F4 buncis berpolong ungu didapatkan informasi bahwa galur PQxGK-1, PQxGI-169, dan GIxPQ-35 telah seragam dalam peubah tipe pertumbuhan merambat, warna batang ungu, warna bunga ungu, dan warna polong ungu tua (Permatasari, 2014). Penelitian ini menggunakan 11 galur buncis ungu (Phaseolus vulgaris L.) F5 hasil persilangan varietas lokal dan varietas introduksi. Dengan demikian penelitian pada generasi F5 ini dimaksudkan untuk memperoleh informasi mengenai keseragaman karakter di dalam galur-galur F5 yang dilihat berdasarkan fenotip tanaman. Penelitian ini bertujuan untuk mengetahui penampilan galur buncis (Phaseolus vulgaris L.) F5 hasil persilangan varietas introduksi dan varietas lokal. Hipotesis yang dapat diajukan ialah diduga dari beberapa galur buncis (Phaseolus vulgaris L.) F5 memiliki penampilan yang sudah seragam untuk beberapa peubah tanaman. Penelitian dilaksanakan pada bulan Januari sampai Mei 2015 di Dusun Suwaluan, Desa Tawang Argo, Kecamatan Karangploso, Kabupaten Malang, Jawa Timur. Alat yang digunakan dalam penelitian ini ialah cangkul, ajir bambu, label, penggaris, timbangan analitik, jangka sorong, kamera, meteran ukur, alat tulis, RHS Colour Chart, panduan pengematan, dan peralatan penunjang penelitian lainnya. Bahan yang digunakan ialah benih dari 11 galur F5 buncis polong ungu hasil persilangan tanaman buncis varietas introduksi dengan varietas lokal, tetua dari 11 galur tersebut, pupuk urea, pupuk SP-36, pupuk KCl, dan pestisida. Penelitian disusun tanpa menggunakan rancangan percobaan dan metode pengamatan berupa Single Plant. Variabel pengamatan dilakukan pada karakter kuantitatif dan karakter kualitatif. Karakter kuantitatif meliputi umur berbunga (hst), jumlah bunga per tanaman, umur awal panen (hst), jumlah polong per tanaman, bobot polong (g), bobot polong per tanaman/hasil (g/tan), panjang polong (cm), lebar polong (cm), dan rata-rata jumlah biji per polong. Pada karakter kualitatif yang diamati ialah tipe pertumbuhan, warna klorofil daun, warna batang, pewarnaan antosianin pada daun, warna dasar polong, corak warna sekunder polong, intensitas warna dasar polong, keberadaan warna sekunder polong, warna standard bunga, warna sayap bunga, tekstur permukaan polong, dan warna utama biji. Analisis statistik yang dilakukan dalam bentuk dendogam ii menggunakan program Numerical Taxonomy and Multivariate Analysis Arithmatic (NTSYSPC-2.02) dan estimasi koefisien keragaman fenotip (KKF) serta koefisien keragaman genetik (KKG). Hasil penelitian menunjukkan bahwa galur PQxGI-169-1-14, PQxGK-1-12-29, GIxPQ-12-2-18, GIxPQ-35-11-23 telah menunjukkan keseragaman dalam semua karakter kualitatif. Galur PQxGI-169-1-14, PQxGK-1-12-29, GIxPQ-12-2-18, GIxPQ-35-11-23, GIxPQ-23-10-39, GIxPQ-19-10-16, GKxCS-6-6-47, GKxCS-108-1-1 dan GKxCS-54-11-44 memiliki keragaman genetik dan keragaman fenotip dengan variabilitas sempit pada semua karakter kuantitatif. Berdasarkan dendogram kemiripan diketahui bahwa semua galur buncis berpolong ungu generasi F5 sudah seragam dalam karakter kualitatif dan karakter kuantitatif yang diamati, karena memiliki derajat kemiripan lebih dari 70%.

English Abstract

Common bean (Phaseolus vulgaris L.) is one of popular commodity in the community. The common bean can be effective in preventing and treating Diabetes Mellitus (Amin, 2014). The statistical data in 2011, 2012, and 2013 was showed that the production of common bean had fluctuated, from 334.659 tonnes, 322.145 tonnes to 327.378 tonnes (Anonymous, 2014). It is feared that does not meet of consumption needs. The effort to improve the quality and production of common bean is made a new variety by crossing between the introduced variety (Purple Queen) with local varieties (Gogo Yellow, Gilik Ijo, Mantili). The crossing purpose is to obtain a new variety that has high yield and purple pod color. In the F4 generation of common bean obtained information that the PQxGK-1, PQxGI-169, and GIxPQ-35 lines had been similar/uniform in growth type, purple stem color, purple flower color, and purple pods variables (Permatasari, 2014). This research use 11 lines of purple common beans (Phaseolus vulgaris L.) F5 as result of crossing between local varieties and introduced variety. Thereby, in the F5 generation is purposed to obtain information of the uniformity on the characters in F5 lines that are seen based on the phenotypic and to find out the appearance of common bean (Phaseolus vulgaris L.) F5 lines. The hypothesis is there are several lines of common bean (Phaseolus vulgaris L.) F5 that have been a uniform performance for several variables. The research was conducted on Suwaluan hamlet, Tawang Argo village, Karangploso subdistrict, Malang, East Java. The tools were used in the research were hoes, bamboo stakes, labels, rulers, analytical scales, calipers, camera, measuring meter, stationery, RHS Colour Chart and equipment supporting other research. The materials were seeds of 11 lines of purple common beans (Phaseolus vulgaris L.) F5, parental of 11 lines common bean, urea fertilizer, SP-36 fertilizer, KCl fertilizer and pesticides. The research was arranged without any experimental design and the observation method was single plant method. The observation variables were quantitative and qualitative character. Data were recorded for quantitative characters like day to maturity, days to flowering, number of flower, number of pods per plant (N pod/pl), weight of pods per plant (g pod/pl), weight of pods per pod (g/pd), pod length (cm pod), pod width (cm pod), average number of seeds per plant (N seeds/pl). The qualitative characters were growth type, chorophyll leaf color, anthocyanin coloration of leaf, stem color, color of standard flower, color of wing flower, ground pod color, secondary pod color, hue of secondary iv pod color, intensity of ground pod color, texture of pod surface, and main seed color. Data were analysed by numerical taxonomy techniques, using NTSYS-pc package, version 2.02. An Unweighted Pair-Group Method of the Arithmetic (UPGMA) average clustering procedure was used to construct dendogram and estimation of genotypic and phenotypic coefficient of variation. The results showed that PQxGI-169-1-14, PQxGK-1-12-29, GIxPQ-12-2-18, GIxPQ-35-11-23 lines had shown uniformity in all the qualitative character. PQxGI-169-1-14, PQxGK-1-12-29, GIxPQ-12-2-18, GIxPQ-35-11-23, GIxPQ-23-10-39, GIxPQ-19-10-16, GKxCS -6-6-47, GKxCS-108-1-1 and GKxCS-54-11-44 lines had genetic coefficient variabition and phenotype coefficient variation with narrow category in all quantitative characters. Based on the dendogram of 11 lines can be known that all lines had been uniform in qualitative characters and quantitative characters were observed, because it has the similarity degree more than 70%.
