Indonesian Abstract

Odd eye ditandai dengan salah satu mata kucing berwarna biru dan mata lainnya berwarna hijau atau oranye. Warna mata dipengaruhi oleh sintesis dan distribusi melanin pada iris. Perbedaan warna mata diduga akibat pengaruh berbagai gen pigmentasi salah satunya adalah gen Tyrosinase. Tyrosinase (TYR) merupakan gen memberikan instruksi untuk membuat sebuah enzim yang dinamakan tyrosinase. Enzim ini terletak di melanosit, yang berfungsi mengatur biosintesis melanin. Melanosit merupakan sel yang menghasilkan melanin, dan melanin merupakan bahan yang memberikan warna pada mata, kulit, dan rambut. Penelitian ini bertujuan untuk menganalisa gen TYR berdasarkan sekuen gen pada kucing. Gen TYR diisolasi dari sampel darah dan diamplifikasi menggunakan metode PCR dengan Primer TYR_F 5’-CATCACGCTGTCCAAAGCTC-3’ dan TYR_R 5’-ATTTCAGACCCTCCAAGCA-3’. Sekuensing dilakukan dengan menggunakan metode Sanger. Analisa sekuen gen dilakukan dengan menggunakan software Sequence Scanner, software MEGA, software BioEdit, dan NCBI BLAST untuk mengetahui homologi sekuen gen TYR pada kucing odd eye, dan kucing non odd eye. Hasil penelitian menunjukkan terdapat perbedaan sekuen DNA pada urutan 664 sampel kucing odd eye terhadap sampel kucing non odd eye dan sekuen gen TYR Felis catus dari NCBI, urutan sekuen ke 664 yaitu basa sitosin (C) menjadi timin (T) (c.664 C>T) dengan mutasi transisi melalui efek silent mutation, dan tidak merubah susunan asam amino, dimana kodon GTC menjadi GTT tetap menyandi asam amino valin (V). Kesimpulan dari penelitian ini adalah terdapat perbedaan sekuen nukleotida gen TYR, namun tidak berdampak pada susunan asam amino.

English Abstract

An odd eye is marked by one of the cat’s eye coloured blue and the other is either green or orange. Eye colour is affected by the synthesis and distribution of melanin in iris. The difference of eye colour is suspected because of the influence of pigmentation genes, one of them is Tyrosinase gene. Tyrosinase is a gene that provides instruction for making an enzym called tyrosinase. This enzyme is located in melanocytes, which can controls melanin biosynthesis. Melanocytes is a cell that creates melanin, and melanin is a material that gives colour to eye, skin, and hair. The aim of this study is to analyze TYR gene based on gene sequence in feline. TYR gene is isolated from the blood sample and amplified using PCR method with primers TYR_F 5’-CATCACGCTGTCCAAAGCTC-3’ dan TYR_R 5’-ATTTCAGACCCTCCAAGCA -3’. Sequencing was performed with the same primer using Sanger method. The gene sequence was analysed using Sequence Scanner software, MEGA software, Bioedit software and NCBI BLAST to determine the homology of TYR sequence gene in the odd-eyed feline and non-odd-eyed feline. The research result showed there is difference between DNA sequence on 664th of odd-eyed feline sample to an odd-eyed feline and TYR gene sequence of Felis catus from NCBI, the 664th sequenced is cytosine (C) base changes to thymine (T) (c.664 C>T) with transition mutation through silent mutation effect and it doesn’t influence the change of amino acid sequence, where GTC codon becomes GTT is still coding valine (V) amino acid. This research can be concluded that although there is sequence difference in TYR gene but it has no effects on protein sequence.
