Indonesian Abstract

Kanker mammae merupakan jenis kanker yang sering terjadi pada anjing betina berumur lebih dari enam tahun. Zat karsinogen DMBA seringkali digunakan dalam pembuatan hewan model kanker mammae. Curcumin dan vitamin E merupakan jenis tanaman herbal yang sering digunakan dalam terapi pengobatan kanker. Curcumin dan vitamin E mengandung zat antioksidan untuk menghambat pertumbuhan sel kanker. Pada penelitian ini bertujuan untuk mengetahui pengaruh kombinasi curcumin dengan vitamin E terhadap ekspresi gen p53 dan gambaran histopatologi kelenjar mammae tikus model kanker mammae akibat paparan DMBA. Penelitian ini menggunakan tikus (Rattus norvegicus) strain Sprague Dawley (SD) yang dibagi dalam 5 kelompok yaitu kontrol negatif (KN), kontrol positif (KP), terapi dosis satu (P1) 48 mg/kgBB curcumin dengan 300 IU/ekor vitamin E, terapi dosis dua (P2) 72 mg/kgBB curcumin dengan 200 IU/ekor vitamin E dan terapi dosis tiga (P3) 108 mg/kgBB curcumin dengan 100 IU/ekor vitamin E. Pada kelompok KP, P1, P2, P3 dilakukan induksi DMBA dengan dosis 10 mg/kg BB sebanyak 10 kali secara subcutan setiap 2 hari sekali dan induksi estrogen dengan dosis 20.000/kgBB sebanyak 5 kali secara intramuscular setiap 4 hari sekali sebelum terapi. Ekspresi p53 diamati menggunakan metode imunohistokimia sedangkan gambaran histopatologi mammae dengan pewarnaan HE. Analisa data ekspresi p53 diamati secara statistik kuantitatif, sedangkan gambaran histopatologi mammae diamati secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa terapi kuratif curcumin dan vitamin E mampu meningkatkan ekspresi p53 hingga 15x. Sedangkan hasil yang ditunjukkan pada gambaran histopatologi mammae, curcumin dan vitamin E mampu memperbaiki gambaran histopatologi mammae berupa penurunan proliferasi sel kanker.

English Abstract

Mammary cancer is the most common type of cancer on female dogs at six years old age. Mammary cancer rat model is frequently induced by carcinogenic substance such as DMBA. Herbal medicines such as curcumin and vitamin E have been developing as a treatment for mammary cancer. Curcumin and vitamin E contain antioxidant substrate to inhibit the growth of cancer cells. This research was determined to study about the effect of curcumin and vitamin E combination on p53 expression and mammary histopathology at rat (Rattus norvegicus) induced by DMBA. This research used Sprague Dawley (SD) strain rat which were separated into 5 groups, negative control (KN), positive control (KP), first therapy (P1) with curcumin dose 48 mg/kgBW and vitamin E dose 300 IU/head, second therapy (P2) with curcumin dose 72 mg/kgBW and vitamin E dose 200 IU/head, third therapy (P3) with curcumin dose 108 mg/kgBW and vitamin E dose 100 IU/head. Positive control group and all therapy groups induced by DMBA at dose 10 mg/kgBW 10x subcutaneously and estrogen induction at dose 20.000 IU/kgBW 5x intramuscularly prior to therapy. The expression of p53 was observed by immunohistochemistry method, whereas mammary histopathology was observed by HE color staining. The data of p53 expression was analyzed quantitatively whereas mammary histopathology was analyzed qualitatively. The result showed that the most effective curative therapy combination of curcumin and vitamin E could increase of p53 expression until 15x. Whereas, the result on mammary histopathology showed that the therapy combination could fixed the histopathology of mammae as the decreasing of cancer cells proliferation.
