Indonesian Abstract

Diabetes Melitus (DM) merupakan penyakit metabolisme ditandai dengan adanya hiperglikemia yang mengakibatkan komplikasi diabetes nefropati sehingga menyebabkan perbaikan struktur ginjal. Kedelai hitam (Glycine max (L.) Merr.) adalah tanaman yang memiliki bioaktif antosianin yang dapat mengobati diabetes melitus. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kedelai hitam terhadap kadar malondialdehid (MDA) dan gambaran histopatologi ginjal tikus (Rattus norvegicus) diabetes melitus. Penyiapan tikus model DM dilakukan dengan injeksi (MLD-STZ) selama 5 hari dengan dosis 20 mg/kg BB. Hewan coba dibagi dalam lima kelompok yaitu kelompok (1) tikus kontrol, kelompok (2) tikus diabetes melitus, kelompok (3) terapi 500 mg/kg BB, kelompok (4) terapi 750 mg/kg BB dan kelompok (5) terapi 1000 mg/kg BB. Pada penelitian ini dilakukan pengukuran kadar malondialdehid dengan menggunakan metode Thiobarbituric Acid (TBA) dan data dianalisis statistik dengan uji Analysis Of Variance (ANOVA) dan dilanjutkan uji tukey. Histopatologi ginjal dengan pewarnaan HE (Hematoksilin- Eosin) diamati menggunakan mikroskop. Hasil penelitian menunjukkan perbedaan nyata kadar MDA antar perlakuan (p<0,05). Pemberian ekstrak air kedelai hitam dosis 1000 mg/kg merupakan dosis optimal yang menurunkan kadar MDA sebesar 42,7%. Pemberian terapi ekstrak dapat memperbaiki histopatologi sel glomerolus mendekati normal. Kesimpulan dari penelitian ini adalah ekstrak air kedelai hitam berpotensi sebagai bahan terapi DM ditinjau dari penurunan MDA dan perbaikan histopatologi ginjal.

English Abstract

Diabetes mellitus (DM) is a metabolic disease characterized by hyperglycemia resulting in complications of neuropath diabetes and damaging of renal structure. Black soybean (Glycine max (L.) Merr.) is a plant that has anthocyanin bioactive to treat diabetes mellitus. This research aimed to study the influences of black soybean water extract to malondialdehyde levels (MDA) and histopathological of renal diabetic rat (Rattus norvegicus). The preparation of DM rat model had been done with injection of (MLD-STZ) for 5 days with a dose of 20 mg/kg. The rats were divided into five groups; group (1) were control rat, group (2) were diabetic rat, and group (3) were the therapy of 500 mg/kg BW, group (4) were the therapy of 750 mg/kg BW, and group (5) were therapy of 1000 mg/kg BW. MDA level was measured by using Thiobarbituric Acid (TBA) method and the data statistics was analyzed with Analysis Of Variance (ANOVA) and tukey test. Renal histopathology with HE staining (Hematoxylin-Eosin) was observed microscopically. The results showed that MDA levels significant different between treatments (p<0.05). Therapy of black soybean water extract dose of 1000 mg/kg was optimal dose to decrease MDA level to be 42.7%. Administering of black soybean water extract therapy could repair histopathological glomerolus cells approaching control. The conclusion of this study was the black soybean water extract can be used as DM therapy based on decreasing of MDA level and repairing of renal histopathological appearance.
