Indonesian Abstract

Hiperkolesterolemia merupakan gangguan metabolisme dimana konsentrasi kolesterol dalam darah melebihi normal. Yogurt susu kambing mengandung bakteri asam laktat, biopeptida, antioksidan, dan vitamin yang dapat menurunkan kadar kolesterol dalam darah. Tujuan penelitian ini adalah untuk mengetahui pemberian yoghurt susu kambing secara preventif dalam mencegah peningkatan kadar Malondialdehida (MDA) dan mencegah peningkatan ekspresi Tumor Necrosis Factor-α (TNF-α) pada organ hati hewan model tikus (Rattus norvegicus) hiperkolesterolemia. Penelitian ini menggunakan 20 ekor tikus jantan umur 10-12 minggu yang dibagi dalam 5 kelompok yaitu kelompok kontrol sehat, hiperkolesterolemia dan dosis pencegahan 300, 600, dan 900 mg/kg BB. Pemberian yoghurt susu kambing diberikan selama 42 hari. Pembuatan hewan model hiperkolesterolemia dengan diet mengandung kuning telur puyuh rebus, minyak babi, dan asam kolat selama 14 hari. Kadar MDA diukur dengan metode Thiobarbituric Acid Reactivity test (TBA-RT) dan ekspresi TNF-α dengan metode imunohistokimia dan dianalisis menggunakan software Axio Vision. Hasil dari penelitian ini menunjukkan bahwa pemberian yogurt susu kambing secara preventif signifikan (p<0,05) menurunkan kadar MDA dan ekspresi TNF-? organ hati tikus antar perlakuan. Pemberian yogurt susu kambing sebagai tindakan preventif dengan dosis 900 mg/kg BB merupakan dosis yang paling efektif untuk menurunkan kadar MDA dan dosis 600 mg/kg BB merupakan dosis paling efektif untuk menurunkan ekspresi TNF- ? organ hati tikus hiperkolesterolemia. Kesimpulan dari penelitian ini, yogurt susu kambing yang diberikan secara preventif dapat digunakan untuk mencegah peningkatan kadar Malondialdehida (MDA) dan mencegah peningkatan ekspresi Tumor Necrosis Factor-α (TNF-α) pada organ hati hewan model tikus (Rattus norvegicus) hiperkolesterolemia.

English Abstract

Hypercholesterolemia is condition when the level of cholesterol in a blood exceed normal level. Goat milk Yogurt contains lactic acid bacteria, biopeptide, antioxidants, and vitamins that can lower cholesterol levels in the blood. The purpose of this research is to know the induction of a goats milk yogurt preventive were able to prevent increased levels of Malondialdehida (MDA) prevent an increase in the expression of Tumor Necrosis Factor-α (TNF-α) and in an animal model of liver organs of rats (Rattus norvegicus) hiperkolesterolemia. This research used 20 male rats, aged of 10-12 weeks which were divided in to 5 groups: control, hypercholesterolemia and preventive doses of 300, 600 and 900 mg/kg body weight. The goat milk yogurt was conducted for 42 days by force feeding method. The hypercholesterolemic rats prepared by administered the egg yolk of boiled quail, lard and cholid acid pigs for 14 days. MDA level was measuring using the Thiobarbituric Acid Reactivity Test (TBA-RT) and the expression of TNF-α by immunohistochemistry methods using Axio Vision software. The result showed that the average of MDA level and TNF-α expression from each group was high significantly different (p < 0.05). Induction of goat’s milk yogurt preventive doses of 900 mg/kg body weight is the most effective dose due to decrease the levels of MDA and the expression of TNF-α hiperkolesterolemia rat liver organs. The conclusions of this research, goat’s milk yogurt preventive were able to prevent increased levels of Malondialdehida (MDA) and prevent an increase in the expression of Tumor Necrosis Factor-α (TNF-α) in an animal model of liver organs of rats (Rattus norvegicus) Hypercholesterolemia.
