Indonesian Abstract

Konsumsi makanan yang mengandung kolesterol tinggi dapat menyebabkan kondisi hiperkolesterolemia. Kondisi hiperkolesterolemia akan ditandai dengan peningkatan LDL. Yoghurt susu kambing mengandung bakteri asam laktat yang dapat mengasimilasi kolesterol dan mendekonjugasi asam empedu untuk menjaga kadar kolesterol darah agar tetap dalam batas normal. Penelitian ini bertujuan untuk mengetahui pengaruh yoghurt susu kambing sebagai pencegahan hiperkolesterolemia terhadap kadar Malondialdehida (MDA) dan ekspresi Tumor Necrosis Factor (TNF-α) jantung tikus. Kadar MDA ditentukan dengan uji TBA dan ekspresi TNF-α melalui metode pewarnaan imunohistokimia. Penelitian ini menggunakan 20 ekor tikus Rattus norvegicus , strain w istar yang berumur 10-12 minggu dengan berat 150-200g, dibagi dalam 5 kelompok yaitu kelompok kontro l , kelompok hiperkolesterolemia , kelompok pencegahan hiperkolesterolemia dengan yoghurt susu kambing dosis 300mg/kg BB +diet hiperkolesterolemia , dosis 600mg/kg BB +diet hiperkolesterolemia dan dosis 900mg/kg +diet hiperkolesterolemia. Pada kelompok pencegahan hiperkolesterolemia, yoghurt susu kambing diberikan terlebih dahulu selama 28 hari kemudian diberi yoghurt dan diet hiperkolesterolemia selama 14 hari. Pada kelompok hiperkolesterolemia diberikan diet hiperkolesterol selama 14 hari. Hasil penelitian menunjukkan bahwa yoghurt susu kambing berpengaruh terhadap kadar MDA, pada dosis 300mg/kgBB terjadi penurunan sebesar 36%, dosis 600mg/kgBB sebesar 60% dan dosis 900mg/kgBB sebesar 73%. Yoghurt susu kambing juga berpengaruh ekspresi TNF-α, terdapat penurunan rata-rata ekspresi TNF-α pada dosis 300mg/kgBB sebesar 24%, dosis 600mg/kgBB sebesar 37% dan dosis 900mg/kgBB sebesar 68%, dari data tersebut dapat disimpulkan bahwa setiap dosis memberikan efek penurunan yang sangat nyata pada kadar MDA dan ekspresi TNF-α (P<0,01). Pada kelompok pencegahan dengan yoghurt mampu memberikan hasil yang berbeda sangat signifikan terhadap kelompok tikus hiperkolesterolemia.

English Abstract

Goat milk yogurt contains lactit acid bacteria that can assimilate cholesterol and deconjugate bile acid to keep the blood cholesterol level normal. The purpose of this research is to examine the effect of goat milk yogurt treatment as Hypercholesterolemia prevention toward Malondialdehyde level (MDA) and the expression of Tumor Necrosis Factor (TNF-α) of rats’ heart. MDA level was obtained by using TBA test, while the expression of TNF-α using immunohistochemistry. This study used 20 Wistar strain rats (Rattus Norvegicus), age 10-12 weeks, weight 150-200 gram divided into 5 groups. They were control group, hypercholesterolemia group, hypercholesterolemia prevention group using 300mg/kgBB, 600mg/kgBB and 900mg/kgBB of goat milk yogurt + hypercholesterolemia diet. In hypercholesterolemia prevention group, yogurt was given 28 days then continued by giving yogurt and hypercholesterolemia diet for 14 days. Meanwhile, hypercholesterolemia group was given hypercholesterolemia diet for 14 days. The result showed that goat milk yogurt does have an effect toward the level of MDA. The using of 300mg/kgBB dosage decreases the level of MDA 36%, 600mg/kgBB dosage decreases MDA level 60% and 900mg/kgBB dosage decreases the MDA level 73%. In addition, goat milk yogurt also affects the TNF-α expression. It was revealed that there was a decreasing of TNF-α expression into 24% for 300mg/kgBB dosage, 37% decreasing of TNF-α for 600mg/kgBB dosage and 68% decreasing of TNF-α expression for 900mg/kgBB dosage. Thus, it can be concluded that each dosage can significantly decrease MDA level and TNF-α expression (P<0.01). The hypercholesterolemia prevention group can show results that very significant by hypercholesterolemia group.
