Indonesian Abstract

Diabetes melitus tipe 1 (DMT1) merupakan kelainan defisiensi insulin akibat kerusakan pankreas yang ditandai oleh hiperglikemia kronis. Pembuatan hewan model diabetes melitus tipe 1 menggunakan Streptozotocin (STZ) yang dapat menyebabkan peningkatan kadar radikal bebas dan inflamasi pada organ pankreas yang kemudian merusak sel-sel β. Curcuma longa L merupakan tumbuhan yang mengandung kurkumin yang bersifat antioksidan dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak Curcuma longa L terhadap kadar IL-6 serta gambaran histologi pankreas pada tikus model DMT1. Penelitian ini bersifat eksperimental dengan menggunakan lima kelompok tikus yaitu kelompok kontrol negatif, kontrol positif, dan tiga kelompok perlakuan (dosis 1,2 gram/kg BB, 1,8 gram/kg BB, dan 2,7 gram/kg BB). Metode penelitian menggunakan Posttest Only Group Design dengan Rancangan Acak Lengkap. Titer Interleukin-6 (IL-6) diukur denganELISA Assay Kit dan gambaran histologi pankreas dengan pewarnaan Hematoksilin Eosin (HE). Analisa data titer IL-6 menggunakan One-Way ANOVA (α = 0,05) serta gambaran histologi pankreas secara deskriptif. Hasil penelitian menunjukkan terjadinya penurunan kadar IL-6. Pengamatan preparat histologi pankreas menunjukkan terjadi penghambatan kerusakan pulau-pulau Langerhans. Simpulan dari penelitian ini yaitu pemberian ekstrak Curcuma longa L dapat menurunkan kadar IL-6 serta menghambat kerusakan pankreas pada tikus model DMT1.

English Abstract

Diabetes mellitus is a deficiency of insulin disorder caused by pancreas damage that marked by chronic hyperglicemia. Diabetes mellitus type 1 animal model was made by induction of Streptozotocin (STZ) that can cause increasing of free radical level and inflammation in pancreas organ that lead to β cell damage. Curcuma longa L is a plant that contains curcumin that has antioxidant and antiinflammation in it. The purpose of this research was to determine effect of Curcuma longa L extract on level of IL-6 and pancreas histology image in diabetes melitus type 1 rat model. This research is experimental research, rats were divided into five groups which were negative control group, positive control group, and three groups of therapy (dosage 1,2 gram/kg weight, 1,8 gram/kg weight, and 2,7 gram/kg weight). This research usesPosttest Only Group Designmethod by Completely Randomized Design. Level of IL-6 was assesed by ELISA Assay Kit and pancreas histology image was stained byHematoksilin Eosin (HE) staining. Analysis of IL-6 level usesOne Way ANOVA (α = 0,05) and pancreas histology image used descriptive analysis. The result shows that level of IL-6 decreases. The pancreas histology image shows that islet of Langerhans damage was blocked. In conclusion, Curcuma longa L decreases IL-6 level and islet of Langerhans damage in diabetes mellitus type 1 rat model.
