Indonesian Abstract

Infeksi enveloped virus, seperti feline immunodeficiency virus banyak menyerang kucing dan dapat menimbulkan gangguan kesehatan yang serius serta kematian. Golongan virus tersebut mudah menghindar dari sistem pertahanan tubuh. Feline tetherin/BST-2 berpotensi untuk dikembangkan sebagai novel antiviral therapy karena dapat menghambat pelepasan berbagai jenis enveloped virus dari sel hospes terinfeksi. Tetherin/BST-2 merupakan protein antiviral yang dihasilkan oleh berbagai jenis sel, seperti PBMCs karena induksi IFN-α. Penelitian ini bertujuan untuk mengetahui tingkat ekspresi mRNA feline tetherin/BST-2 pada PBMCs yang diinduksi IFN-α. PBMCs berasal dari darah kucing. PBMCs yang dikultur terbagi menjadi kelompok kontrol/tanpa perlakuan, serta kelompok perlakuan A dan B yang masing-masing diinduksi dengan IFN-α dosis 100 U/ml dan 1000 U/ml selama 24 jam. Kuantifikasi ekspresi mRNA feline tetherin/BST-2 dilakukan dengan real-time RT-PCR. Data output dianalisa secara kuantitatif deskriptif. Hasil penelitian menunjukkan bahwa nilai rata-rata ΔCT kelompok perlakuan lebih kecil dari pada kelompok kontrol. Kesimpulannya, pemberian IFN-α mampu menginduksi ekspresi mRNA feline tetherin/BST-2 pada PBMCs yang ditandai dengan menurunnya nilai ΔCT pada kelompok perlakuan dibanding dengan kelompok kontrol.

English Abstract

Enveloped virus infection, such as feline immunodeficiency virus, is one of the common infection of cats causing serious health impairments and death. This virus tend to evade immune system. Feline tetherin/BST-2 has recently been recognized as potent antiviral protein that inhibits the release of enveloped virus particles from infected host cells. Tetherin/BST-2 is widely expressed in response to IFN-α, including PBMCs. This research was aimed to evaluate the expression level of feline tetherin/BST-2 mRNA in PBMCs induced by IFN-α. PBMCs obtained from venous blood of a cat were cultured into untreated (control) and treated groups. The treated groups were induced with 100 U/ml IFN-α (treated group A) and 1000 U/ml IFN-α (treated group B) for 24 hours. Expression level of mRNA feline tetherin/BST-2 was quantified by real-time Reverse Transcriptase-Polymerase Chain Reaction (real-time RT-PCR). Output data were analyzed descriptively. This study showed that average of treated group’s ΔCT value was lower than untreated group’s. It proves that IFN-α was able to induce feline tetherin/BST-2 mRNA expression.
