Indonesian Abstract

Salmonella spp. merupakan salah satu mikroorganisme yang dapat menyebabkan foodborne disease pada produk ayam broiler yang dapat mengkontaminasi sejak dari peternakan. Kurangnya penerapan biosekuriti pada lingkungan peternakan dapat menyebabkan transmisi dan kontaminasi Salmonella spp. Penelitian ini bertujuan untuk mengetahui adanya cemaran Salmonella spp. serta mengetahui karakteristik Salmonella spp. di peternakan ayam Mertojoyo dan Sawojajar. Sampel yang digunakan adalah feses, pakan dan air minum yang berasal dari peternakan ayam di Mertojoyo dan Sawojajar, Malang. Penelitian ini dilakukan melalui tahapan metode mikrobiologi standar, uji biokimia, uji serologis dan Sodium Dodecyl Sulphate Polyacrilamid Gel Electrophoresis (SDS-PAGE) untuk mengetahui profil pita protein Salmonella spp. dari sampel peternakan ayam broiler. Hasil karakterisasi bakteri berdasarkan uji mikrobiologi, uji biokimia, uji serologi dan profil pita protein dianalisa secara deskriptif. Hasil isolasi bakteri dari feses di Mertojoyo ditemukan kisaran jumlah bakteri dengan rerata 4,9x108 ±11,34 cfu/ml dan Sawojajar 3,2x108 ±1,70 cfu/ml, sedangkan isolasi bakteri dari air minum dengan kisaran jumlah rerata, Mertojoyo 5,8x108±16,36 cfu/ml dan Sawojajar 3,8x108 ±4,82 cfu/ml. Dari 5 isolat hasil karakterisasi fenotipe berhasil dilakukan pendugaan isolat mendekati genus Salmonella choleraesuis, Salmonella sub genus I (S. kauffmannii; S. enterica; S. enteritidis), Salmonella sub genus II (S.salamae; S.dar-es-salaam), Salmonella sub genus IV (S. houtenae). Hasil analisis SDS-PAGE menunjukkan bahwa isolat F25A dan F25B mempunyai pita protein yang sama dengan berat molekul 36,29 kDa yang diduga sebagai S. enteritidis atau S. typhimurium. Sehingga berdasarkan hasil penelitian ini dapat disimpulkan adanya cemaran Salmonella spp. di lingkungan peternakan ayam dan diketahui adanya keanekaragaman Salmonella spp.

English Abstract

Salmonella spp. is one of microorganism that caused foodborne disease in broiler poultry products which can contaminate since in the poultry farm. Salmonella spp. transmission and contamination caused from less of implementation of biosecurity. The aim of this research was to analyze the presence and characteristic of Salmonella spp. in poultry farm. Faecal, feed, and water samples that were used in this research taken from poultry farm in Mertojoyo and Sawojajar, Malang. This research was conducted by standard microbiological method, biochemistry assay, serological assay, and SDS-PAGE to determine the protein profile of Salmonella spp. The result of bacteria characterization based on microbiological test, biochemistry assay, serological assay, and protein profile were analyzed descriptively and variation of Salmonella spp. were determined based on protein profile. The isolation result showed that there were Salmonella spp. contamination in faecal were, Mertojoyo 4,9x108 ±11,34 cfu/ml and Sawojajar 3,2x108 ±1,70 cfu/ml and in drink water was, Mertojoyo 5,8x108±16,36 cfu/ml and Sawojajar 3,8x108 ±4,82 cfu/ml. From phenotypic characterization of 5 isolates could be predicted that the isolates have similiarity with Salmonella choleraesuis, Salmonella sub genus I (S. kauffmannii; S. enterica; S. enteritidis), Salmonella sub genus II (S.salamae; S.dar-es-salaam), Salmonella sub genus IV (S. houtenae). Based on the result of SDS PAGE showed that the F25A and F25B isolates has a functional protein bands with a molecular weight of 36,29 kDa which suspected as S. enteriditis and S. typhimurium protein. As the summary of this research, there were presenced and diversity of Salmonella spp. in poultry farm.
